Objective: To study the role of T-bet[a T helper 1(Th1)lymphocyte transcription factor]gene expression in aplastic anemia and evaluate the correlation between T-bet/IFN-γ.signal transduction and severe aplastic anemia.Methods: Thirty-six patients and fifteen healthy volunteers entered this study. The expression of T-bet gene was examined by real-time quantitative polymerase chain reaction (RT-QPCR) in peripheral blood mononuclear cells collected from patients and healthy controls. The concentration of IFN-γin the serum was determined with enzyme-1inked immunosorbent assay(ELISA).Results: The expression level of T-bet in SAA patients was increased compared with that of CAA patients(P=0.001)and healthy controls(P﹤0.001).There was no significant diference between CAA patients and healthy controls(P=0.528). In the SAA group,the concentration of IFN-γin the serum was significantly elevated compared with that in the CAA patients(P﹤0.001)and healthy controls(P﹤0.001). There was no significant diference between CAA patients and healthy controls(P=0.325).Conclusions: T-bet/IFN-γsignal transduction involved in the pathophysiology of severe aplastic anemia.
