Objective To study the growth inhibiting and apoptosis induced by sodium phenylacetate in human breast cancer cell line MCF-7 and to approach its mechanisms.Methods To deal exponential phase of growth human breast cancer cell line MCF-7 with different density sodium phenylacetate 72 hours, using MTT assay to detect absorbance(OD) value, calculating inhibition ratio. The apoptotic ratio and the cell cycle was analyzed by flow cytometry. The expression of Bcl-2 and Bax protein were assessed by immunocytochemistry, and the expression of p21WAF1/CIP1 mRNA was determined by reverse transcription-polymerase chain reaction (RT-RCR).Results The inhibition ratio of human breast cancer cell line MCF-7 increased with density multiplication of sodium phenylacetate. The inhibition ratio was the maximum when the density was 15mmol/L which was 48.14%. Apoptotic peak appeared in DNA bargraph of flow cytometry. Quantitative analysis result of flow cytometry was that the cell apoptotic ratio of 3, 9, 15mmol/L density was 4.16%, 9.05%,11.12%. Compared with control group, the difference was significance(P<0.05).With the density of sodium phenylacetate increased, the cells in G0/G1 stage multed gradually, while S stage descended gradually. Immunocytochemical methods illustrated that the expression of the protein of Bax obviously ascended in contrast with that of the contrast group cells while the expression of the protein of Bcl-2 obviously descended in contrast with that of the contrast group cells. Expression of p21WAF1/CIP1 mRNA was up-regulated through ladder strap as the increasing of sodium phenylacetate density.Conclusion Sodium phenylacetate has conspicuous growth inhibiting effect and inducing apoptosis on human breast cancer cell line MCF-7.The apoptosis mechanisms probably was concerned with Bcl-2 gene expression down-regulation and Bax,p21WAF1/CIP1 gene expression up-regulation.
