Abjective To study on differentiation of rat bone marrow mesenchymal stem cells into neuron-like cells .METHODSRat bone marrow mesenchymal stem cells were obtained and were isolated and cultured. They were randomly divided into control group , medium from cultured olfactory ensheathing cell group as well as GM1 group and then cells were induced to culture. Inverted phase contrast microscope was used to observe the growth and morphological changes of primary cells and passage cells .Immunohistee hemcial method was adopted to detect.The specific labeled neural nest protein of neura1(Nestin) . the specific label of neural cells, neuron specific enolase(NSE),β-tubulin .RESULTS:(1) Primary cells:24 hours later,1ots of cells adhered to the wall and were in big ancipital fibroblasts with clear borderline between caryon and cytoplasm,and most of them were moneearyons. As the time increasing,most of cells grew in settlement with rapid proliferation and different size of colony. Cells in settlement were typical fusiform cells and the colony were close to each other, 80% of which mixed together at one week later and were in whirlpool arrangement.(2) Passage cells: cells were round and in floating status. which adhered to the wall rapidly and evenly arranged.Compared with primary cells,the shapes of passage cells were in unified fusiform shape.(3) Determination of the specific label of nerve cells:little expression of neural nest protein(Nestin) was positive in the control group .1hour,6hours after induction,positive expressions of Nestin were found in the GM1 group group.5 hours,24--48 hours later, positive expressions of NSEand B—tubulin were found. In certain time,the intensity of expression of above-mentioned indicators were gradually increased.CONCLUSION: BMSCs can be amplified and purified in short time and be induced into neuronal-like cell by medium from cultured olfactory ensheathing cell and GM1。...
