Objective:To observe protective effects of the donors pretreated with NF-κB inhibtor PGA1 on donors' heart and to study effects of which on the function of recipient's T lymphocytes in rat model of cardiac transplantation.Methods:In this experiment,24 albino Wistar and 24 Sprague-Dawley rats were, respectively,selected as donors and recipients.A model of rat heterotopic heart allograft was established by using Ono's technique.Experimental animals were divided randomly into two groups:experiment and control groups.Experiment group:donors were pretreated with PGA1 by intraperitoneal injection at the dose of 20nmol/kg each day for 3 days before transplantation.And,donors of control group pretreated with vehiculum.After transplantation,six rats of each group were used observing survival time of donor' heart,and others were sacrificed on the 5thday to analyze the function of T lymphocytes from recipient.The histopathological examination of allograft tissues were performed,mixed lymphocyte culture(MLC)were done by MTT method, levels of serum IL-2 and IL-4 were determined by enzyme-linked immunosobnent assay(ELISA).Continuous variables are presented as the mean±SD;comparisons between groups were performed with Student's t test;differences were considered to be statistically significant at value of p<0.05.Results:(1)the survival time of cardiac allograft in experiment group was prolonged compared with that of control group.(9.2±1.2d vs 6.3±0.5d,p<0.05).(2) the rejection of cardiac allograft in experiment group was moderated compared with that of control group.(3)MLC showed that proliferative responses of spleen cells to alloantigen stimulation from experiment group of recipients was suppressed compared that of control group.(0.52±0.03 vs 0.67±0.05,p<0.05).(4)the serum IL-2 level was reduced in experiment group compared that of control,(133±12pg/ml vs 175±14pg/ml, p<0.05);there was no statistically significant difference between two group.(43±7pg/ml vs 47±8pg/ml,p>0.05).Conclusion:(1)by donor pretreated with the NF-κB inhibitor PGA1,the survival time of cardiac allograft was prolonged.(2)by donor pretreated with the NF-κB inhibitor PGA1,the proliferative responses of T lymphocytes to alloantigen stimulation of recipients was suppressed;the serum IL-2 level of recipients was decreased but the IL-4 level of those was unchanged;T lymphocytes populations in allograft was reduced. (3)PGA1 can suppressed the function of T lymphocytes of the recipients,thus inhibited allograft rejection and prolonged the survival time of cardiac allograft,probably this effect of PGA1 come from that it inhibited the activity of NF-κB of donors.
