Objective The breast cancer is one of the most common malignant minors in recent years. And the past,the incidence of breast cancer is rising year by year,which threatening the health of women.To research and explore the pathogens of breast cancer is of great significance.It is presumed that the risk factors of breast cancer are related to high estrogen exposure levels,oncogene activation and suppressor gene inactivation. At the same time,the abnormality of some metabolic enzymes still possible participate in the development of breast cancer.GSTP1,encoding theπ-class GSTs supergene family,is a kind of detoxicate enzyme,which playing an important role in the process of degradation and evacuation to blastomogen.The distinction of structure,function and expression in GSTP1 gene can lead to different capability toresist blastomogen, which causing different susceptibility in individual.In this study,we detected the mutation and polymorphism in GSTP1 gene for all exons to analysis the relationship with breast cancer and to find the effect of gene structure changes of GSTP1 in the tumorigenesis of breast cancer.Methods Specimens were from Yunnan Province,which included 50 cases of breast cancer,50 cases of adjacent hyperplastic tissues,50 cases of benign hyperplasia tissues,50 cases of normal breast tissues and 15 cases of normal blood lymphocytes cells.HE chips of the specimens were diagnosed by WHO standard.PCR-SSCP and DNA sequencing methods were used in this study.Results(1)There were 3/50 cases(6%)mutations of GSTP1 gene in exon3 and 5/50 cases(10%)mutation of GSTP1 gene in exon4 in breast cancers,we found no mutations in any of the exons in all of the adjacent hyperplastic tissuesl,benign hyperplasia tissues and normal breast tissues.(2)We detected single nucleotide polymorphism(SNP)in exon5,which included 12/50 cases(24%)in breast cancer tissues,5/50 cases(10%)in adjacent hyperplastic tissues,3/50 cases(6%)in benign hyperplasia tissues,5/50 cases(10%)in normal tissues,1/15 cases(6.7%)in normal lymphocutes.(3)Statistical analysis showed that,the age of patients,the type of organization,the clinical stage and whether metastasis have nothing to do with GSTP1 gene mutation(P>0.05).The polymorphism frequency of GSTP1 gene in exon 5 was discrepant in cancer,precancerous lesions and normal crowd(P<0.05),the highest frequency was found in cancer cases.Conclusions(1)The mutation of GSTP1 gene may be partly related to the occurrence of breast cancer;(2)The hot mutations of GSTP1 gene are exon3 and exon4;(3)People with GSTP1 genes polymorphism may be susceptible to breast cancer risk in Yunnan Province;(4)The age of patients,the type of organization,the clinical stage and whether metastasis have nothing to do with GSTP1 gene mutation;(5)The structure abnormity of GSTP1 is various.It is helpful to further illustrate the relationship between gene mutation and expression for GSTP1 gene and the function of GSTP1 gene in the tumorigenesis and the development of breast cancer.
