[Objective]To investigate the expression and its significance of BMI-1 gene in leukemia patients。[Methods]BMI-1 gene level was assessed by reverse transcription polymerase chain reaction(RT-PCR)in 34 acute leukemia patients' bone marrow mononuclear cells(BMMNC),26 chronic myeloid leukemia (CML)patients,6 chronic lymphoblastic leukemia(CLL)patients, 10 normal controls,as well as leukemia cell line K562。The regularity of the expression of BMI-1 gene in leukemia and the relationship between BMI-1 level and the prognosis of leukemia was explored。[Results]The expression of BMI-1 gene was positive in leukemia cell line K562,but negative in 10 controls as well as CLL。The BMI-1 gene was expressed in 15.4%(n=4)CML patients and 47.1%(n=16) AL patients。The expression ratio of BMI-1 gene in CML-CR(5%)was significantly lower than that in CML-BP(50%)(P<0.05),and The expression ratio of BMI-1 gene between AML(57.7%)and ALL(12.9%) has statistical difference(P<0.05),while the expression ratio between CML-CR and AL has no statistical difference(P>0.05)。The expression of BMI-1 gene positive in newly diagnosed leukemia changed to negative after complete remission,but the reverse transmition reoccurred after relapse。[Conclusions]BMI-1 gene was overexpressed in leukemia patients, and may be related to curative effect of disease。BMI-1 gene expression may be involved in the occurrence and development process of leukemia, it may be used as a molecular marker for leukemia treatment targeting the provision of new sites。...
