Objective:â‘ To understand the distribution of HLA-B27 gene in healthy population from Yunnan Dai Ethnic Group,by polymerase chain reaction-sequence specific primers(PCR-SSP),HLA-B27 gene was amplified from the healthy Dai inhabiting in Xishuangbanna Autonomous Prefecture in Yunnan Province,that will provide important references for racial genetics.â‘¡Using polymerase chain reaction-sequence specific primers to amplify HLA-B27 gene in patients with leukemia in Yunnan Han people and study the association of HLA-B27 gene with these patients,that provide some reference in genetics for the pathogenesis of leukemia and provide the basic data for the relevance of HLA to different ethnic groups with leukemia in Yunnan for study of the next step.Methods:â‘ A total of 61 healthy unrelated individuals from Dai ethnic groups lived in Xishuangbanna Autonomous Prefecture in yunnan province of southwestern China were enrolled in this study.The peripheral blood was collected after informed consents were obtained,and then the HLA-B27 gene was amplified by PCR -SSP.Using four-frame Table X~2 examination to analysis differences of HLA-B27 gene frequencies in healthy unrelated individuals from Yunnan Dai and Han,Naxi,Lisu,Nu,Yi,the South Han,the North Han in China and Thai people in Thailand.â‘¡26 patients with leukemia were diagnosised in the second hospital of Kunming Medical College,between in May 2007 and November 2007,who were selected randomly from the department of Hematology.62 Yunnan Han Who lived in Yunnan province of southwestern China were enrolled as the control.After extrating the genomic DNA from peripheral blood anticoagulanted by EDTA and then the HLA-27 gene was amplified by PCR-SSP.According to case-control study analysis,we analysised the difference of HLA-B27 gene frequencies between in the healthy control group and the Leukemia Group and studied the association of HLA-B27 gene with patients with leukemia.Result:â‘ 61 cases in Yunnan Dai healthy population for HLA-B27 gene,14 cases were positive,accounting for 22.9%,significantly higher than that of Yunnan Han(4.8%),Naxi(0.6%),Lisu(0.9%),Nu(0%),Yi(4.2%),China Southern Han(4.3%),the North Han(4.29%)and Thai people(10%).Using the four-frame table X~2 test, studied the distribution of HLA-B27 gene frequencies among Yunnan Dai,Han,Naxi,Lisu,Nu,Yi,Southern Han,North Han in China and Thai people.There are significantly different to frequencies of HLA-B27 gene among these people.â‘¡In 26 cases of patients with leukemia in Yunnan about HLA-B27gene research, we found one cases is positive,for 3.8 percent,and in 62 cases Han people,three cases is positive,accounting for 4.8%.According to case-control study on the analysis method,there was no statistically different between the patients with leukemia and the control subject.Conclusion:â‘ In the study about,we found that frequence of HLA-B27 gene in healthy people from Yunnan Dai ethnic group was significantly higher than that of the Han,Naxi,Lisu,Nu,Yi,and also significantly higher than Southern Han,the North Han in China and Thai people in Thailand.The results of this study show that, Xishuangbanna Dai Han is a different group with a high frequency of HLA-B27 gene comparing with other groups.May be due to different regions,different ethnic groups and the distribution of HLA itself that there is a big difference,or be due to living environment where different groups of people lived,living habits of different groups of people and the different experimental methods,and so on,that result in the distribution of HLA-B27 gene frequency uneven.Hence need to exclude affecting factors as much as possible and need to collect a larger sample and go to further reduce the error.â‘¡In the study of this group to the relevance of HLA-B27 gene and the patients with leukemia in Yunnan Han,we didn't find that there were the relevance of HLA-B27 gene and the patients with leukemia,and also didn't find that there were difference between in the patients with leukemia and in healthy people to the distribution of HLA-B27 gene frequencies.This may be related to the incidence of leukemia with complicated mechanism,more classification and geographical distribution of the samples,sample size,and so on.Need to collect large numbers of samples and study association of different types of leukemia from different nationalities with HLA.
