Objective to establish a method for culture of rat and human adipocyte, to better understand hyperplasia and hypertrophy properties of human adipose tissue.Methods Using "tissue culture" and "collagenase digest" strategy, fibrobiast-like cell were cultured from SD rats and adult human abdominal adipose tissue.the morphological changes of the culture cell observed and the growth curve drawn. The intracytoplasmic lipid of the culture cells was determined using oil red 0 staining.Results the cultured fibroblast-like cells showed highly homogeneous appearance with active proliferation ,and could differentiate into mature adipocytes.oil O staining and morphological and biological observation verified these cells as preadipocytes with accumulation of lipid droplets in the cytoplasm.Conclusion predipocytes are present in mature human and SD rats adipose tissue and possess the potential to proliferate and differentiate into mature adipocytes. These homogeneous preadipocytes with good proliferateing activity can be harvested through primary cell culture. This study laid the basis for further probing into obesity. Objective To investigate the relationship between the single nucleotide poly -morphism in adipokine Apelin gene and type 2 diabetes mellitus in Chinese from Kunming.Methods A total of 329 unrelated Chinese subjects (216 patients with type 2 diabetes and 113 non-diabetic control individuals without family history of diabetes) from Kunming were screened for the presence of the 7250A→G mutation in the Apelin gene by use of mispairing base—nested polymerase chain reaction --restriction fragment length polymorphism.Results There were no significant differerces in the frequencies of Apelin gene 7250A→G polymorphism between the type 2 diabetes mellitus and control groups(P=0.421,P>0.05).Conclusion The apelin gene 7250A→G polymorphism is not associated with type 2 diabetes mellitus in Chinese from Kunming.
