Objective Cholesteryl ester transfer protein(CETP)plays an important role in lipoprotein metabolism.It facilitates the transfer of cholesteryl ester from high-density lipoprotein(HDL)to apolipoprotein(Apo)B-containing lipoproteins.CETP gene is known to have many single nucleotide polymorphisms which have been associated with CETP activity.Several studies have reported that the CETP-TaqIB gene polymorphism is associated with HDL-C levels and the risk of coronary artery disease,but the results are inconsistent.There are fifty-six ethnic groups in China.Han is the largest group and Zhuang is the largest minority.Geographically and linguistically,Zhuang can be classified into 43 ethnic subgroups,among which Hei Yi Zhuang is proved to be the most conservative subgroup.The population size is about 52,000.Little is known about the effect of the CETP-TaqIB gene polymorphism on serum lipid levels in this population.Threrfore,the present study was undertaken to compare the difference in the CETP-TaqIB gene polymorphism and its association with the serum lipid levels between the Guangxi Hei Yi Zhuang and Han populations.Methods A total of 758 subjects of Hei Yi Zhuang aged 15-70 were surveyed by a stratified randomized cluster sampling.Information on demographics,socioeconomic status,cigarette smoking,alcohol consumption, and physical activity was collected with standardized questionnaires.Blood pressure,height,weight,serum lipid and apoliporotein levels were measured, and body mass index(BMI)was calculated as a measure of weight relative to height.Gene polymorphism was determined by polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP)combined with gel electrophoresis,and then confirmed by direct sequencing.The results were compared with those in 778 matched subjects of Han Chinese from the same region.Results(1)Serum levels of total cholesterol(TC),triglycerides(TG), low-density lipoprotein cholesterol(LDL-C)and ApoB were significantly lower in Hei Yi Zhuang than in Han subjects(P<0.01 for all),whereas the levels of HDL-C and the ratio of Apo AI to Apo B were significantly higher in Hei Yi Zhuang than in Han(P<0.01 for each).There was no significant difference in Apo AI levels between the both ethnic groups(P>0.05).(2)The frequencies of B1 and B2 alleles were 65.2%and 34.8%in Hei Yi Zhuang,and 63.2%and 36.8%in Han(P>0.05),respectively.The frequencies of B1B1,B1B2 and B2B2 genotypes were 43.0%,44.3%and 12.7%in Hei Yi Zhuang,and 41.7%,45.1% and 13.3%in Han(P>0.05),respectively.(3)The levels of HDL-C and Apo AI in Hei Yi Zhuang were higher in B2B2 genotype than in B1B1 genotype(P<0.05),whereas the levels of TG and Apo AI in Han were higher in B1B2 or B2B2 genotype than in B1B1 genotype(P<0.01).No difference in the remaining lipid parameters was found among the three genotypes in the both erhnic groups(P>0.05 for all).(4)Spearman rank correlation analysis showed that the CETP-TaqIB genotypes were significantly correlated with HDL-C and ApoAI levels in Hei Yi Zhuang(P<0.01 and<0.05),but there were no significantly correlated with serum lipid parameters in Han(P>0.05 for all). Multivariate logistic regression analysis also showed that the levels of HDL-C, LDL-C and Apo AI were correlated with age,sex,alcohol consumption and cigarette smoking(P<0.01~0.05).TC and ApoB levels were correlated with age,BMI and ethnic group(P<0.01 for all).TG and LDL-C levels were correlated with BMI and cigarette smoking(P<0.01 for all).HDL-C and ApoB levels were correlated with ethnic group and sex(P<0.01~0.05)in the combined population of Hei Yi Zhuang and Han.Conclusions There were no significant differences in the allelic and genotypic frequencies of CETP-TaqIB polymorphism between the Hei Yi Zhuang and Han pupolations,but B2 allele of the CETP-TaqIB was correlated with high HDL-C and Apo AI levels in the Hei Yi Zhuang and high TG and Apo AI levels in the Han population.
