A Study Of Cross-Resistance Between Antibiotics In Pseudomonas Aeruginosa In Vitro

Objective:To found induced model of antibiotic-resistance with Pseudomonas aeruginosa wild-type PAO1 in vitro,and observe the crossresistance of the induced model with the other antibiotics,to provide theory about the selection of antibiotics in therapy for the infection caused by Pseudomonas aeruginosa.Methods:First,to carry out a antibiotics susceptibility test which aim at non-fermentative bacilli with Kirby-Bauer Method,and then choose susceptible antibacterials to induce PAO1 resistant to them by the Method of increase subinhibitory concentration by degrees,and accompany a control group of in-synchronism,monitoring the sensitivity of PAO1 to the antibiotics that recommended by NCCLS in antibiotics susceptibility test of Kirby-Bauer Method to non-fermentative bacilli,note the diameter of the ring that the PAO1 can not grow in it,then analyze these diameters by SPSS13.0 software.Results:1,Wild-type PAO1 susceptible to all antibiotics that use in antibiotics susceptibility test except for Bactrim and Tetracycline.2,Choose susceptible antibiotics of Sulperazon,Levofloxacin and Amikacin to induce PAO1 resistant to them with the Method of increase subinhibitory concentration by degrees,can set up induced model of Sulperazon-resistance,induced model of Levofloxacin-resistance and induced model of Amikacin-resistanc,at the same time,we observed that: (1)After induced PAO1 resistant to Sulperazon,PAO1 can confer cross-resistance to antibiotics of carbapenems:Tienam and Meropenem;to antibiotics of fluoroquinolones:Levofloxacin and to beta-lactam antibiotics: Aztreonam.And not found cross-resistance to other antibiotics that PAO1 susceptible to them before induce with Sulperazon.The PAO1 of control group of in-synchronism have the same consequenc of antibiotics susceptibility test as the PAO1's that before induce.(2)After induced PAO1 resistant to Levofloxacin,PAO1 can confer cross-resistance to antibiotics of fluoroquinolones:Ciprofloxacin and to antibiotics of carbapenems:Tienam and Meropenem.And not found cross-resistance to other antibiotics that PAO1 susceptible to them before induce with Levofloxacin.The PAO1 of control group of in-synchronism have the same consequenc of antibiotics susceptibility test as the PAO1's that before induce.(3)After induced PAO1 resistant to Amikacin,PAO1 can confer cross-resistance to aminoglycoside antibiotics:Gentamicin.And not found cross-resistance to other antibiotics that PAO1 susceptible to them before induce with Amikacin.The PAO1 of control group of in-synchronism have the same consequenc of antibiotics susceptibility test as the PAO1's that before induce.Conclusion:1,Wild-type PAO1 resistant to Bactrim and Tetracycline,susceptible to cephalosporins antibiotics:Ceftazidime,Sulperazon and Cefepime;susceptible to beta-lactam antibiotics:Piperacillin,Piperacillin/Tazobactam and Aztreonam; susceptible to carbapenems antibiotics:Tienam and Meropenem;susceptible to aminoglycoside antibiotics:Gentamicin and Amikacin and susceptible to antibiotics of fluoroquinolones:Ciprofloxacin and Levofloxacin before induced with antibacterials. 2,Wild-type PAO1 can be induced to resistant to Sulperazon,Levofloxacin and Amikacin,and Wild-type PAO1 resistant to Levofloxacin is easier than to Sulperazon and Amikacin.3,After Wild-type PAO1 resistant to Sulperazon by induce,PAO1 can confer cross-resistance to Tienam,Meropenem,Aztreonam and Levofloxacin.4,After Wild-type PAO1 resistant to Levofloxacin by induce,PAO1 can confer cross-resistance to Ciprofloxacin,Tienam,and Meropenem.5,After Wild-type PAO1 resistant to Amikacin by induce,PAO1 can confer cross-resistance to Gentamicin.
