Effect Of Gingko Bilobate Extract On Function And Proliferation Of Cultured Peripheral Blood Lymphocyte From Patients With Systemic Scleroderma

Objectives To investigate the effect of Gingko bilobate extract(GBE) on cytokines secretion and proliferation of cultured peripheral blood lymphocytes from patients with systemic scleroderma(SSc).Methods Peripheral blood lymphocytes from patients with systemic scleroderma and healthy adult were cultured in vitro,lymphocyte growth rate were counted and excretion level of cytokine IL-4 andγ-IFN were measured 60 hours after adding GBE,dexamethasone injection(DEX)and cyclophosphamide injection(CTX)in different concentration.Results①25g/L GBE had no significant effect on proliferation of cultured peripheral blood lymphocytes from patients with SSc.50 g/L GBE show evident acceleration effect on proliferation,however GBE acceleration effect on lymphocyte proliferation descended in 100g/L.The acceleration effect was concerned with GBE concentration.②Both DEX and CTX could inhibit peripheral blood lymphocyte proliferation from patients with SSc cultured in vitro.③IL-4 secretary quantity in all treatment groups had no statistically significant difference compared with that in blank control group.④γ-IFN secretary quantity was concerned with intervened drugs and their concentration. Both 100g/L and 50g/L GBE could increaseγ-IFN secretion (P=0.000,P=0.019),but 25g/L GBE had no distinct effect onγ-IFN secretion(P=0.866).DEX had no significant effect onγ-IFN secretion(P=0.093). 0.13g/L CTX could increaseγ-IFN secretion(P=0.017).⑤GBE,DEX and CTX had no significant effect on IL-4 andγ-IFN secretion and proliferation of cultured peripheral blood lymphocytes in healthy adults control groups.Conclusions GBE could accelerate peripheral blood lymphocyte from patients with SSc proliferation in vitro and its effect was concerned with concentration.Both 100g/L and 50g/L GBE could increaseγ-IFN secretion. DEX could inhibit peripheral blood lymphocyte from patients with SSc proliferation but had no effect on IL-4 andγ-IFN secretion.CTX could inhibit peripheral blood lymphocyte from patients with SSc proliferation,0.13g/L CTX could increaseγ-IFN secretion.
