Objective:To investigate the therapeutic effect of radiotherapy in combination with hyperthermia on RD cells of rhabdomyosarcoma. Comparing the therapeutic effects of different orders of hyperthermia and radiotherapy, in order to provide the experimental support for clinical treatment of rhabdomyosarcoma.Methods:The experiment was divided into five groups: contrast, radiotherapy, hyperthermia,radiotherapy after hyperthermia, and hyperthermia after radiotherapy. RD cells were exposed to radiation, heating, radiation combined with hyperthermia respectively in vitro. Counting the cells number and the colony number,drawing the growth curve,calculating the rate of the colony formation of different groups on day 14.Drawing the change of the cell period distribution and calculating the apoptosis rate of different groups. Results:1. There was significant difference of growth inhibitory effect between radiotherapy combined with hyperthermia and radiation or hyperthermia alone(p<0.05). There was significant difference of growth inhibitory effect between radiotherapy after hyperthermia and other groups(p<0.05).2. The rate of the colony formation were 1.4%, 4.7% for radiotherapy combined with hyperthermia groups and were 13.0%, 6.7% for radiation or hyperthermia alone group. There was significant difference of rate of the colony formation between radiotherapy combined with hyperthermia and radiation or hyperthermia alone(p<0.05). There was significant difference of rate of the colony formation between radiotherapy after hyperthermia and other groups(p<0.05).3. There was significant difference of rate of accumulation of cells at stage G2+M between hyperthermia alone and other groups(p<0.05).4. There was significant difference of apoptosis rate between radiotherapy combined with hyperthermia and radiation or hyperthermia alone(p<0.05). There was significant difference of apoptosis rate between radiotherapy after hyperthermia and other groups(p<0.05).Conclusions:1. The growth inhibitory effect of radiotherapy combined with hyperthermia was significantly better than radiation or hyperthermia alone .Radiotherapy after hyperthermia was better than other groups.2. Hyperthermia alone can block RD cells at stage G2+M.3. The killing effect of radiotherapy combined with hyperthermia was significantly better than radiation or hyperthermia alone.4. The killing effect of radiotherapy after hyperthermia was better than other groups.
