The Expression Of TNF-α And IL-6 In The Liver On Oleic Acid Induced ALI Rat

Objective:To observe the expression of TNF-αand IL-6 in the liver on oleic acid induced ALI rats.To discuss the relevance of cytokine and the liver injury during ARDS by oleic acid and to provide experimental basis on further control the secondary by ARDS and MODS.Methods:sixteen healthy Wistar rats were randomly divided into control group,OA6h group.The oleic acid was adopted to build ALl model in OA6h group from caudal vein(0.15mg/kg),control group was injected normal sodium(0.15mg/kg) from caudal vein.Liver function(ALT,AST,TB) of each group was detected,The W/D of tung was measured.The expression of TNF-α,IL-10 protein in hepatic tissue of each group was detected by radioimmunoassay.The expression of TNF-α,IL-6mRNA in hepatic tissue of each group was detected by RT-PCR.The structure of liver was observed by light microscope.Results:Compared with control group,W/D and liver function of OA6h group increased(P＜0.01).The content of TNF-αand IL-6 protein in blood plasma and hepatic tissue increased compared with control group(P＜0.01).The expression of TNF-αand IL-6mRNA in hepatic tissue increased compared with control group(P＜0.01). Conclusions:Functional disturbance of liver may take place during ARDS by oleic acid, the expression of TNF-αand IL-6 was increased in gene and protein level,TNF-αand IL-6 may play a role in the function disturbance of the liver during ARDS.
