ObjectiveTo investigate the effect of vascular endothelial growth factor in hepatic ischemia/reperfusion injury and the function of anti-vascular endothelial growth factor monoclonal antibody(anti-VEGF mAb)in alleviating hepatic ischemia/ reperfusion injury.MethodsThirty normal male Sprague-Dawley rats were randomly divided into three groups.Namely group C that was subjected to ischemia/reperfusion injury with intraperitoneal administration of anti-VEGF mAb(100ug/300g)15min before ischemia;group B with the same injury followed by saline administration in the same manner,and group A with only anesthetization leparotomy but not ischemia.Animals were killed at 6 hours after reperfusion.Routine assays were performed for testing the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),lactate dehydrogenase(LDH),the activity of hepatic myeloperoxidase(MPO)and the hepatocyte apoptosis.The expression of VEGF in the liver was examined by Western blot and the hepatic pathological changes were evaluated in hematoxylin and eosin (HE)stained sections.ResultsThe levels of serum ALT,AST,LDH and the MPO activity in hepatic tissues increased after hepatic ischemia/reperfusion injury(all P<0.01).The expression of VEGF protein was also increased significantly.But these effects were offset by administration of anti-VEGF mAb(P<0.01).In group B,widespread pathological changes and hepatocyte apoptosis were observed in the hepatic tissue following hepatic ischemia/reperfusion injury,while similar changes were scarcely visible in groupe C duo to the protective effect of intraperitoneal administration of anti-VEGF mAb 15min before ischemia. ConclusionThe up-regulation of VEGF protein in hepatic ischemia/reperfusion injury is involved in the process of liver injury.Hepatic ischemia/reperfusion injury can be alleviated by anti-VEGF mAb treatment.
