AIMTo investigate the regularity of hepatocyte apoptosis and the effect of restraining kupffer cell to the hepatocyte apoptosis and the expressiong of related genes in liver ischemia reperfusion(I/R)injury in rats.METHODS100 male Wistar rats(180-220 g,7-8weeks age)were divided into two groups at random:Gdc13 exposure group(Gd)and ischemia/reperfusion group(IR).In Gd group,inject Gdc13 via the tail vein in the first two days(10mg/kg);the IR group inject the same mount of isotonic Na chloridethe.The third day,left branches of portal vein, hepatic artery,hepatic duct were blocked up for 60 min and then opened to establish liver I/R models in rats;In each group,samples were collected in 0.5,1,6,12 and 24h after reperfusion respectively.Serum alanine ALT,AST by auto biochemical analyzer, and TNF-αby ELISA,and the level of MDA,SOD,GSH in liver was measured,the incidence of hepatocyt apoptosis was measured by TUNEL,immunohistochemical straining of survivin and caspase-3 was determined with optical microscope,the morphology changes were observed by EM.RESULTSThe level of ALT in each time,AST measured in 6,12h time after reperfusion in Gd groups were significantly lower than that in IR groups.The level of TNF-αin the Gd groups in all the time were lower than that in the IR groups.The level of MDA measured in 6,12,24h after reperfusion in Gd groups was totally significantly lower than that in IR groups,and the level of SOD in 12,24h,the level of GSH in each time after reperfusion in Gd groups were higher than that in IR groups(~ap<0.05).The express level of surviving(IOD)measured in each time after reperfusion in Gd groups were totally significantly higher than that in IR groups corresponding(~ap<0.05);the express level of TUNEL and Caspase-3(IOD)measured in 0.5,1,6,12,24h time after reperfusion in Gd groups were totally significantly lower than that in IR groups corresponding(~ap<0.05).CONCLUSIONRestraining KC may decrease the hapetocyte apoptosis,than improve the liver function.It may react through increasing the expression of survivin and down regulating the expression of caspase-3.
