Effects Of Tumor Necrosis Factor-α, Pioglitazone And Fenofibrate On TIMP-1 Expression In 3T3-L1 Adipocytes

Objective To observe the effects of tumor necrosis Factor-α,pioglitazone and fenofibrate on TIMP-1 mRNA expression in 3T3-L1 adipocytes.Methods Treat the 3T3-L1 adipocytes with TNF-α,pioglitazone and fenofibrate under diversity concentration and diversity phase,using the RT-PCR to measure the TIMP-1 mRNA expression level.Results(1) In 3T3-L1 preadipocyte,the expression level of TIMP-1 mRNA was decreased,With the differentiation from 3T3-L1 preadipocytes to 3T3-L1 adipocytes, the expression level of was decreased from 0 day to 4 days,was increased from 4 days to 8 days.(2) 0.1,1,10,50ng/ml of TNF-αtreated in 3T3-L1 adipocyte for 24 hours, the relative TIMP-1 mRNA values were increased in a dose-dependent manner.50ng/ml of TNF-αtreated in 3T3-L1 cell of undifferentiated,ifferentiating,differentiated TIMP-1 mRNA increased in a time-dependent manner.(3) 0.1,1,10,100 mol/L of pioglitazone treated in 3T3- L1 adipocyte for 24hours,the TIMP-1mRNA values were decreased in a dose-dependent manner.100μmol/L of pioglitazone treated 3T3-L1 cell of undifferentiated,differentiating,differentiated,TIMP-1 mRNA decreased in a time-dependent manner.(4) 0.1,1,10,I00μmol/L of fenofibrate treated in 3T3- L1 adipocyte for 24hours,the TIMP-1mRNA values were decreased in a dose-dependent manner.100μmol/L of fenofibrate treated 3T3-L1 cell of undifferentiated,differentiating,differentiated,TIMP-1 mRNA decreased in a time-dependent manner.(5) While 50ng/ml of TNF-αand 100μmol/L of pioglitazone compared with 50ng/ml of TNF-αtreated 3T3-L1 cell in different stages with different concentrations,the expression of TIMP-1 mRNA decreased.compared with 100μmol/L of pioglitazone,the expression of TIMP-1 mRNA increased.(6) While 100μmol/L of fenofibrate and 100μmol/L of pioglitazone compared with 100μmol/L of fenofibrate treated 3T3-L1 cell in different stages with different concentrations,the expression of TIMP-1 mRNA decreased.compared with 100μmol/L of pioglitazone,the expression of TIMP-1 mRNA decreased.Conclusion TNF-αincreases TIMP-1 mRNA expression in a time-dependent manner in 3T3-L1 cells at different stages and in dose-time-dependent manner in 3T3-L1 adipocytes.PIO and FF decreases TIMP-1 mRNA expression in a time-dependent manner in 3T3-L1 cells at different stages and in dose-time-dependent manner in 3T3-L1 adipocytes.Pioglitazone reverses TNF-αstimulated TIMP-1 mRNA expression in 3T3-L1 cells at different stages.PIO and FF do not have synergistic effects on TIMP-1 expression in 3T3-L1 cells at different stages.
