Fructus Cnidii is the mature fruit of Cnidium monnieri( L.) in umbelliferae families and cnidium cuss genera Cuss. Fructus Cnidii is widely distributed throughout China, mostly in Hebei and Shandong province. The chemical constituents of Fructus Cnidii are composed of coumarin with some physiological activities. The isolation and identification of coumarin from Fructus Cnidii were osthol, isopimpinellin, columbianadin, imperatorin, and so on. Chinese traditional medicine Fructus Cnidii has antifungal, antiviral, anti-ascaris, anti- trichomonal effects and similar effect as prednisone, which has been used Externally and clinically to kill worm, stop pruritus,warming the kidney to strengthen yang. On the other hand, coumarins from Fructus Cnidii has effect on the ovariectomized and osteoporosis in rats, which can also effectively strengthen hormones -induced osteoporotic rats bone density and can prevent and treat osteoporosis, especially postmenopausal osteoporosis.The aim of the dissertation is to study the extraction of coumarin from Fructus Cnidii, techniques of effective part preparation, quality control standards and pharmacodynamics of the osteoporosis. The results are as follows.In the extraction technology study of effective part, the coumarin is extracted from Fructus Cnidii by orthogonal-test-optimized process of advance supercritical CO2 extraction (SFE) technique. The content of total coumarin in extractant is above 50% tested by the ultraviolet spectrophotometric method.In the techniques study of effective part preparation, the active constituents of the forming preparation, coumarins are all fat-soluble compounds which can hardly be dissolved in water, which taste numb, hot and can cause burning stomach sensation after oral administration. Coumarins are mostly light-sensitive compounds. In order to develop the scope of the clinical application and make theoretical basis for suitable forms of prepared drugs,the experiment made the total coumarins into inclusion complex,that is, to be included intoβ-cyclodextrin, which can get capsules and improve the stability and the solubility and cover its taste.In the study on quality standards for capsules, the capsules are identified by TLC, and HPLC is applied to determine the components of the total coumarins in the capsules. It is proved that the latter method is accuracy, repeatability, and high reproducible. The dissolution characteristics of osthole and imperatorin is also tested to control the reagent's qualityOn the base of the research above, we study pharmacodynamics on osteoporosis. The results of experiment show that coumarin capsules can enhance maximum tolerance of rat's femur, resistance to lateral bend, flexural modulus, bending fracture of energy and bending energy. It can also thicken bony cortex, improve morphology of bone-trabecula and increase the number of bone-trabecula, average width of bone-trabecula and percentage of bone-trabecula. It is obvious the preparation has evident therapeutic effect on the osteoporosis.In a word, all the above results provided the reliable theory foundation for the clinical test of drug.
