Partâ… : Expression of Ets-1, MMP-1 and VEGF in Colorectal CarcinomaObjective:To study the expression of E26 transformation-specific (Ets-1), MMP-1 (matrix metalloproteinases-1) and VEGF (vascular endothelial growth factor) in human colorectal carcinoma and explore the role of Ets-1 in angiogenesis and metastasis.Methods:Collect the pathological species of colorectal carcinoma proved by operation during 2003-2004 from Tongji Hospital affiliated by Huazhong University of Science and Technology. By using immunohistochemical staining (SP method), we detected the Expression of Ets-1, MMP-1 and VEGF in colorectal carcinoma (n = 61) and normal colon tissues (n = 12), and then analyze the relationship of them with clinical data.Result: Ets-1, MMP-1 and VEGF were negatively expressed in all normal mucosa tissues. The positive rate of Ets-1, MMP-1 and VEGF expression was 75.4%, 78.7% and 82.0% in colorectal carcinoma respectively. No significant correlation was found between the positive rate of them and tumor's size as well as differentiation (P >0.05). There were significant associations with Duke's stage, the depth of invasion, lymphatic invasion and distant metastasis (P <0.05). Correlation analysis displays Ets-1 expression was positively associated with MMP-1 and VEGF level.Conclusions:Ets-1 was overexpressed in colorectal carcinoma, and related with clinical stage, invasion and metastasis. Ets-1 expression was positively related with MMP-1 and VEGF level. The expression of Ets-1 can become a reference index to predict malignant biology behavior of colorectal carcinoma.Partâ…¡: Expression of Ets-1 in Human Colorectal Carcinoma Cell LinesObjective:To study the correlation of Ets-1 expression with the metastatic potential of human colorectal carcinoma.Methods:Expression of Ets-1 in various cell lines of human colorectal carcinoma possessed with different metastatic potentials (Lovo and HT-29) was investigated by reverse transcription polymerase chain reaction technique(RT-PCR) and immunohistochemical staining (SP method) in mRNA and protein level.And expression of Ets-1 in normal colon tissues was examined normal as control.Results: The Ets-1 mRNA and protein were well expressed in both of Lovo and HT-29 cell lines, but Lovo cells expressed higher than HT-29 cells whether in protein or mRNA level. Ets-1 mRNA and protein were negatively expressed in normal colon tissues.Conclusions:The expression of Ets-1 is related to the metastatic potentiality of human colorectal carcinoma cells.
