Latanoprost is an ophthalmic topica used for glaucoma and ocular hypertension. In the course of clinical application, doctors find that Latanoprost can promote eyelashes and hairs around the eye growing, which was listed as its side effect. Therefore, we presume that a topica for hair growth may be made of Latanoprost. To prove whether it is ture, I designed and carried out this experiment. The objective is to investigate the effect of Latanoprost (LT) on hair growth and explore the mechanism of action, by using it on rats'skin and observing the change of local hairs, and by culturing isolated hair follicles in vitro, comprising of the length of hair growth, proliferation hair follicle cells and morphological transmutation.The study was divided into two parts: preventive experiment investigate the effect of LT on rats'hair growth. 28 adult SD rats were given the local application of LT(in different concentrate) compared with the free form (control group). The density of follicles and dermis blood capillary were observed with microscope.Determine the expression of TGF-β1 by immunohistochemistry and detect its mRNA by reverse transcription polymerase chain reaction (RT-PCR).The second part investigate the effect of LT on human scaple hair growth. Model of culturing isolated hair follicles in vitro was established with Williams E medium. Screen 120 pieces of anagen hairs and divide into six groups: testing group with 0.0001%0.0005%,0.001% and 0.005% latanoprost ,negative control group and positive control with 25ug/ml minoxidil .Observe the morphological changes of hair follicles with invert microscope and measure overall hair lenghth and of the size of individual zigzag hair segments using a eyepiece microscope. Comprehend the proliferation of hair follicle cell and count melanocytes by [3H]Thymidine Labeling.The following results were found in preventive experiment: 1. Compared with control groups, the hair length of growth in higher concentration of experimental groups show great significant difference (P<0.05), and the experimental groups of lower concentration show no difference with control groups, P>0.05. 2. Histological results indicate that amounts and calibers of rat`s dermis blood capillary in all groups don`t show manifest difference, and density of follicles is greater in groups with 0.05%LT and there is no difference in other groups. 3. The expression of TGF-β1 in anagen hair decrease evidently in groups with 0.05%LT than other groups. Culturing human scalpe hair follicles, we known that hair follicles in groups with 25ug/ml minoxidil 0.001% and 0.005%LT grew faster than those in blank control group, and HFs with 0.01%LT grew slowllier than others, and the difference was of statistical significance. Analytic results of 3H-TdR-uptake rate were as same as aboving.Conclusion: The local application of LT can promote rat hair growth and the mechanism of action may be involved in down-regulation of TGF-β1. Latanoprost has the scalp hair growth promoting effect, which react on cultured follicles in the third to seventh day.The effect of latanoprost is closely related with its concentration: lower concentration has promoting effect and higher concentration has inhibiting effect in a certain concentration scope. And the possible mechanism is related with the following aspects: regulating (enhancing or inhibiting) hair follicle growth, lasting or decurtating anagen, making hair follicle cataplasia(telogen) delay or in advance and then effect(promote or inhibit) hair growth.
