Part I the expression of Mahoganoid protein and it's mRNA in the testes and epididymides of mature ratObjective To localize the Mahoganoid protein and Mahoganoid mRNA in the testes and epididymides of mature male rat.Methods Testes and epididymides obtained from mature male SD rats (n=20) were 4%polyformaldehyde -fixed and sliced for immunohistochemistry (IHC) test with an antiserum and in situ hybridization(ISH) test, respectively, for detecting Mahoganoid protein and Mahoganoid mRNA . Results In the 2 tests, Clear immunopositive staining was both observed in the Leydig cells, spermatogoniums, primary spermatocytes, spermatids, Sertoli cells, and peritubular myoid cells. And in epididymal tissues, the Mahoganoid protein and Mahoganoid mRNA are respectively expressed in principal cells, basic cells and epithelial cells. Besides the Mahoganoid protein are mainly located on cell membrane and cytoplasm and Mahoganoid mRNA are mainly located on cell cytoplasm.Conclusion Both Mahoganoid Protein and Mahoganoid mRNA are expressed in the male rat reproductive system and localized in Leydig cells, Sertoli cells, spermatogenic cells and epithelial cells of epididymal tissues etc. It may play an important role in the process of spermatogenisis but its physiological significance remains to be clarified. PartⅡDistribution of Mahoganoid protein and its mRNA in the testes and epididymides of rats in different ages.Objective To localize the Mahoganoid protein and it's mRNA in the testes and epididymides of rats in different ages and study the gene's significance in spermatogenesis.Methods First, the Mahoganoid protein and it's mRNA were detected by immunohistochemical (IHC) test and in situ hybridization (ISH) test, respectively ,in the testicular and epididymal tissues obtained from SD rats (n = 10x5) of different ages(0d,5d,20d,50d,70d). Then photos of each slide were taken and analyzed by medical image analysis system. At last, all experimental data were processed by SPSS13.0 software package. ANOVA was performed.Results The expression of the Mahoganoid protein and its mRNA was showed by brown staining and observed in the Leydig cells, spermatogonia, primary spermatocytes, spermatids, Sertoli cells, and peritubular myoid cells of testicular tissues and principal cells, basic cells and epithelial cells of epididymal tissues. Besides Mahoganoid protein was mainly located within cell membrane and cytoplasm, while it's mRNA was mainly located within cell cytoplasm. Compared with that of the control group, the expression of the Mahoganoid protein and it's mRNA in both the 50d and 70d groups was stronger then in 20d, 5d and 0d groups(P〈0.05). Meanwhile, the expression in 20d groups was stronger then in 5d and 0d groups(P〈0.05〉.Conclusion Both Mahoganoid protein and Mahoganoid mRNA are expressed in the male rats'reproductive system and localized in Leydig cells, Sertoli cells, spermatogenic cells and epithelial cells, etc. Besides the expression of the Mahoganoid protein and it's mRNA increased distinctly with the maturation of sex. The results implies mahogaoid gene may play an important role in spermatogenesis.
