Expression And Significance Of ILK And α-SMA In Experimental Hepatic Fibrosis

Objective: To investigate the expression of integrin-linked kinase(ILK) andα-smooth muscle actin(α-SMA)in mouse experimental hepatic fibrosis and its significance.Methods: Liver fibrosis models of rats were made by injecting with 40% CCl4. Thirty male and thirty female SD rats were randomly divided into five groups and scarified for 2,4,6,8 weeks after injecting of CCl4 respectively. The liver tissues of rats in all groups were stained by hematoxylin-eosin(HE) ,reticular fiber staining,and were employed by immunohistochemical response,and then the pathological changes, fibra accrementition and the expression of ILK andα-smooth muscle actin(α-SMA) in rat liver tissues were observed.Results:Immunohistochemistrically,the signal of ILK was not detected nearly in normal liver tissues,and only slight signal ofα-SMA was detected in normal liver tissues. The expression of ILK andα-SMA in liver tissues of rats after injection with CCl4 were found obviously higher than those in normal rats(P<0.005, P<0.05).Moreover,it present with progressive tendency for the expression of ILK andα-SMA in rats respectively at 2nd,4th,6th and 8th week after injection with CCl4(P<0.05).The signal of ILK andα-SMA were detected mainly in perisinusoidal space of Disse, proliferative portal space,fibrotic septation and cells around hepatic sinusoid near to fibrotic septation.They nearly expressed in the common area.The expression of ILK andα-SMA was enhanced gradually and obviously during the experimental hepatic fibrosis, an positive correlation was found between the expression of ILK andα-SMA ,and an positive correlation was also found between their expression and the state of hepatic fibrosis(P<0.01).Conclusion: There was a close relation between ILK andα-SMA, it also indicated that there was a close relation between ILK and HSC activation.So ILK may be involved in the pathogenesis of hepatic fibrosis, and understanding the mechanism of action of ILK in hepatic fibrosis,which may provide a new theory for the pathogenesis,prevention and cure of hepatic fibrosis...
