| Objective :To explore the effects of Verapamil on hepatocytes apoptosis, expressions of Bax and Bcl-2 in rats with obstructive jaundice.Methods:Ver group and OJ group rats were anesthetized with pentobarbitone by intraperitoneal injection, bile commen duct was cut off after being liberated to erect obstructive jaundice rat model. bile commen ducts of SH group rats were liberated without being cut off.Ver group rats were intraperitoneal injected with Verapamil(10mg/kg·d) after operation,SH group and OJ group were treated with normal sodium.6 rats of every group were killed in 7d and 14d. 2ml venous blood from rats was used to test serum TBil, DBil, ALT and AST.Apoptosis of hepatocytes was tested by TUNEL and accounted apoptosis index(AI). To apply HE staining and the histopathologically hepatic changes were observed by optical microscope. the expressions of Bax and Bcl-2 were detected by immunohistochemistry method.Result:①There was significant difference between AST and ALT of OJ7 group,Ver7 group and SH7 group(P<0.05);②There was significant difference between AST and ALT of OJ14 group,Ver14 group and SH14 group(P<0.05);③The significantly histopathologically hepatic changes were not observed by optical microscope in HE staining of SH group in different time points. The significantly histopathologically hepatic changes were observed of OJ group.The degree of changes in Ver group was between OJ and SH group in the same time point.④A few of hepatocytes apoptosis in SH group rats can be seen in different time points,the number of hepatocytes apoptosis in OJ group obviously increased. the number of hepatocytes apoptosis in Ver group was between OJ and SH group and there was significant difference between three groups (P<0.05);⑤There was low level expression of Bax and Bcl-2 in SH group and there was significantly difference between OJ group and SH group(P<0.05).The expression of Bax in Ver group was lower than OJ group (P<0.05) and the expression of Bax accorded with AI.The expression of Bcl-2 in OJ group and Ver group was higher than SH group(P< 0.05),there was significantly difference between Ver group and OJ group (P<0.05).Conclusion:①Obstructive jaundice can induce hepatocyte apoptosis in rats,Bax play a important role in the control of hepatocyte apoptosis in obstructive jaundice rats;②Verapamil can decrease the hepatocytes apoptosis which induced by obstructive jaundice;③Verapamil may be directly or indirectly enhance the level of anti-apoptosis protein Bcl-2 and decrease the level of apoptosis protein Bax in obstructive jaundice rats. |
