Prostate is one of the most susceptible organs to many diseases and prostate cancer is the main cause of death in USA. Morbidity of prostate cancer lies in the first place in Northern Europe and the second place in USA followed lung cancer. In china, morbidity of prostate cancer increased in recent 20 years and the potential morbidity over 50 years old in autopsy is comparable to that in USA,which is 5%-20%.Many patients miss their best time of surgery because of having no symptoms in early stage. Presently, the main diagnosis methods in prostate cancer are digital rectal examination (DRE) and PSA (prostate special antigen) and prostate puncture, but the result is variable being affected by many other causes. So the accuracy is variable and many patients can't tolerate the pain in prostate puncture. So exploring new valuable,specific exam methods and tumor marker for early prostate cancer diagnosis become very urgent in this field. Our research will test the expression of S100A8 in the tissue of prostate caner by immunohistochemistrical as well as RT-PCR. At the same time contrast the coincidence of DCE-MRI and other clinic diagnostic methods to pathologic staging for early prostate cancer and estimate the value of S100A8 and DCE-MRI in diagnosis. Aim: Estimate the value of S100A8 and DCE-MRI as useful methods in early prostate cancer diagnosis by studying the expression of S100A8 in tissue in prostate cancer and by contrasting the diagnostic results of DCE-MRI and other methods to pathologic staging.Methods: The expression of S100A8 in human normal prostate tissue (NP),benign prostatic hyperplasia (BPH) and prostatic carcinoma (Pca) is examined by SABC way.The expression of S100A8 mRNA in human normal prostate tissue,(NP) benign prostatic hyperplasia (BPH) and prostatic carcinoma (Pca) is examined by RT-PCR.Analyze the pathologic grading results of 42 patients with the diagnostic results got by DRE, MRI and DCE-MRI before surgery; valuate the coincidence of DCE-MRI to early-stage prostate cancer diagnosis.Results: 1.The result of immunohistochemistry: In specimens of normal prostate tissue(NP),the male expression rate is 16.66%(1/6).In specimens of BPH, the positive expression rate is 19.35%(6/31),In specimens of prostatic carcinoma (Pca), positive staining mainly located in cytoplasm and nucleus of cancer cell and positive expression rate is 78.57%(22/28),which is significantly different from normal prostate tissue (NP) and benign prostatic hyperplasia (BPH)(P<0.05). The expression of S100A8 has significant correlation with the pathologic grading as well as clinical stages of prostatic carcinoma (P<0.05).2. The result of RT-PCR: there is no expressions In specimens of normal prostate tissue(NP)and expressed in both specimens of BPH and Pca, while the expression intensity of mRNA is significantly different between BPH and Pca, which is proved by gray scale analysis software Quantity One and SPSS11.0. S100A8 mRNA is expressed weakly in benign prostatic hyperplasia (BPH) but strongly in prostatic carcinoma (Pca).3.Coincidence of DCE-MRI to pathologic grading diagnosis for 42 early-stage prostate cancer contrasted with DRE and MRI: there is no significantly different between DCE-MRI and pathologic staging diagnosis for prostate confined tumor(0.05
