Objectives This study was to investigate the distribution of the gene polymorphism of calpain 10 (CAPN10) SNP-19 in the Han nationality population in Zhenjiang of China, and to assess the relations between the gene polymorphism of CAPN10 SNP-19 and type 2 diabetes mellitus (T2DM) and body weight.Methods A total of 212 unrelated Han people of Zhenjiang was randomly selected. 120 T2DM (According to the diagnostic criteria of WHO in 1999, males, 62; females, 58; average age, 55.72±12.27) and 92 non-diabetes normal controls (without T2DM family history, males, 48; females, 44; average age, 57.54±6.37) were recruited in the Hospital of Jiangsu University. By WHO criteria in 1997, there were 132 non-overweight and obesity (males, 64; females, 68; average age, 57.02±9.41) and 80 overweight and obesity ( males, 46;females, 34; average age, 55.68±11.30) among them. Genomic DNA was extracted from the collected blood samples, using TaKaRa-blood genome DNA extraction kit, and polymerase chain reaction ( PCR )was used to detected the genotype of site SNP-19 in CAPN10 gene. Anthropometric variables, blood glucose, insulin and other clinic parameters were measured.Results Higher FPG, TG, TC, LDL-C, BMI and HOMA-IR were found in the T2DM group (P<0.05), The frequency of SNP-19 genotype and allege was not differed between the T2DM group and non-diabetes group (P>0.05); Higher FINS, WHR, HOMA-IR and lower LDL-C were found in the individuals with genotype 22 than those of individuals with genotype 11 and 12 in T2DM group(P<0.05); Higher FPG, TG, WHR and HOMA-IR were found in the overweight and obesity group(P<0.05); The frequency of 2 allele and 22 genotype of site SNP-19 in the overweight and obesity group (P<0.05) was significantly higher than that in the controls with BMI<25kg/m~2 (P<0.05).Conclusion The SNP-19 of the CAPN10 gene associated with risk factors about diabetes mellitus, CAPN10 gene polymorphism might play some roles in the etiology of T2DM; The SNP-19 genotype 22 in the CAPN10 gene might contribute to the development of overweight and obesity.
