Detecting the distribution of ER-α,ER-βby Western blot and RT-PCR.Setting up the ER-βknock-out mice model of EMs by autotransplantation.Measuring the weight of the lesions and analyzing the difference of weights between ER-β-/-and the control(ER-β+/+)groups.Testing the expression of MMP-9 in lesions, conserved uterus and douche of peritoneal cavity between ER-β-/- and the control (ER-β+/+)group by Elisa and RT-PCR respectively.Testing expression of VEGF by RT-PCR between the ER-β-/- and the control(ER-β+/+)group.Results:1.The distribution of ER-αin EMs and AM:There is no difference between normal,endometria and nomal uterine muscles(P>0.05);as well as the normal endometria and eutopic endometria of EMs(P>0.05).However,there is significant difference between endometrial cysts and normal endometria (P<0.05),endometrial cysts and eutopic endometria of EMs(P<0.05),lesions of AM and nomal uterine muscles(P<0.01).2.The distribution of ER-βin EMs and AM:There is no difference between each two of the five groups(P>0.05).3.There is no difference of the weight of the lesions between the ER-β-/- and +/+ groups.4.The expression of the MMP-9 in the ER-βknockout mice:There is no difference of the expression of the MMP-9 between the ER-β-/- and +/+ lesions and the douche of peritoneal cavity,as well as the uterus transplantation group and uterus transplantation plus ovariotomy group.However,there is significant difference between the lesions and the remaining uterus.5,The expression of the VEGF-A in the ER-βknock-out mice:There is no difference of the expression of the VEGF-A between the ER-β-/- and +/+ lesions,as well as the uterus transplantation group and uterus transplantation plus ovariotomy group.Conclusion:(1)In this study,we demonstrated that the expression of ER-αis much higher in endometrial than in lesions.Unlike the phenotype of ER-α,ER-βshowed an average and normal expression in the detected areas.(2)There is no significant differences of the weights of the lesions,the ability of invasion and angiogenesis between the control and the ER-βknock-out groups although the MMP-9 of transplanted lesions is higher than eutopic uterus.Thus,we infer that ER-βdoes not paly a major role in EMs and may be only an auxiliary factor in the process. | | Keywords/Search Tags: | endometriosis, EMs, adenomyoma, AM, ER-a, ER-β, gene knockout, MMP-9, VEGF, western blot, RT-PCR, Elisa | PDF Full Text Request | Related items |
