Studies On Mechanisms Of Active Monomer, RR7, Extracted From Rhodobryum Roseum Limp. In Protecting Against Myocardial Ischemia

Abstract/Summary:
In this reseach we study the effect of Rhodobryum roseum Limp in protecting against myocardial ischemia.R.roseum is the traditional chinese medicine locatity in Yunnan Province.Many nations in China have used it as a herb.It was recorded that it can help to cure heart disease,some about heartache,flustered,heart-throb,short breath and so on. And preliminary pharmacologic experiments,we find that active compounds in R.roseum play on important role in heart disease,especially RR7 from acetoacetate fraction.In our study,RR7 has shown the antioxidation and relieve myocardial ischemia effect,moreover it has been not detected that animal experiment's application of RR7 had any obviously adverse reaction and toxic action.So to interpret RR7's mechanism of action thoroughly, we use pharmacological and molecular mechanism's method to study RR7's mechanism of action systematically.Antioxidation of RR7 was studied by decapitation experiment in mice and hypoxia under normal pressure in mice.Recently studies showed that RR7 can reinforce the ability of anti-anoxia of mice model,significantly prolonging the survival time of model and protection of myocardial ischemia and cerebral anoxic.Besides,it is depend on the concentration of RR7.We carry out in vivo experiment to study anti-hypoxia affect of RR7.Using the MI model induced by hypophysin to test the effect of antioxidative enzyme level in serum and myocardium of low-dose,middle-dose and high-dose RR7 group.The result showed that the antioxidative enzyme of Propranolol's group,RR7 Groups of high and middle-dosage have significantly increased.Then we establishing the model of cardiac muscle cell induced by isoprel.The conclusion is that the RR7 maybe have apparente antioxidation.And the RR7 have antioxidantive effect in the model of mitochondria of cardiac muscle cell induced by hydrogen peroxide too.The result showed that protection of MI by RR7 probably enhance SOD,GSH,AST and the activity to Na+-K+ ATP,Ca2+ATP.It is also decrease production of MDA.It is can relieve oxidative damage obviously by the mitochondria in cardiac muscle,raising the ability of anti-hypoxia to the cardiac muscle.Recently it shows thatβreceptor blocker can reduce the mortality and relieve arrhythmia,and it is the only one drug being demonstrated which can lower sudden death.First of all,activity preliminary screening by the rats' electrocardiogram monitoring and the histopathology examination induced in ISO,it shows that the heart rate of rats in RR7 groups is obviously lower than model group,and the peak of T wave significantly decrease,suggesting that RR7 may have relieved arrhythmia function.Besides,it is found that the RR7 groups can obviously decrease the impaired cardiac muscle tissue by histopathology examination of the rat's cardiac muscle.It is suggests that necrosis of cardiac muscle fiber and the cardiac muscle cell reduce significantly by electron microscope detection.Studies of RR7 through the mice anti-hypoxia model reflected evident results.The RR7 which can elevate capability of mice anti-hypoxia and prolong the survial time has the protective effect of heart muscle ischemia and cerebral anoxia,and does dependent.The results is that RR7 has the evident effect of anti-hypoxia.As the RR7 has the anti-hypoxia effect,we did the antioxygen experiments in vitro.The observation of ISO induced cardiac muscle cell model reflected that RR7 has the protective effect of the heart muscle ischemia.Use the ISO-induced heart muscle ischemia model,detect the effect of Rhodobryum roseum Limp on the expression ofβacceptor protain,compared with the componds with high,middle and low dose.The adiminstration is lower than the induced ones.In the level of cAMP,the RR7 with middle and high dosage improved the level of cAMP significantly.
