PrefaceIntracerebral hemorrhage is the most common and fatal disease in neurology, it's still a big threat to human health all over the world. So it's very important to get more information about the pathophysiological mechanism of the disease and therefore we can get better therapeutic methods.In the present study,we measured brain Apo-J mRNA around the hemorrhagic lesions in the operated side and non-surgical side of the acute experimental intracerebral hemorrhage rats ,and evaluate whether there were differences in the two sides.Aflter that,we analyzed whether relativities between Apo-J mRNA expressions and brain water contents(BWC),and therefore laying the foundation for exploring the possible pathophysiological mechanism of Apo-J in intracerebral hemorrhage.Methods54 Sprague-Dawley adult rats were randomly devided into normal control group, sham- operated group,intracerebral hemorrhage 3 hours group ,6 hours group, 12 hours group, 1 day group,3 days group,5 days group,7 days group. Intracerebral hemorrhage models were prepared by injecting autologous arterial blood into the right caudate nucleus.Both in the operated sides and the non-surgical sides,around the hemorrhagic lesions or the corresponding positions,The total RNA were isolated with RNAiso Reagent.Meanwhile, the apo-J mRNA expressions in operated sides and non-surgical sides(right sides and left sides for the control group) were analyzed by RT- PCR. Brain water contents (BWC) were measured. We compared the apo-J mRNA expression level differences between the groups,and tried to explore whether there were relations between the expression levels and BWC.SPSS 13.0 statistical software was used to analyze the data.All data were expressed as x|-±s.A value of p<0.05 was considered statistically significant.Results1. The expression of Apo-J mRNA(1) Comparison between groups in the operated sides:①between intracerebral hemorrhage 3d group,control group and sham-operated group, low levels of Apo-J mRNA expressions were obtained in both control group and sham-operated group. There were no differences between the two groups(p>0.05).The Apo-J mRNA expression levels of the intracerebral hemorrhage 3d group were higher than both the control group and the sham-operated group,significant differences were obtained(p<0.001).②Between the intracerebral hemorrhage groups,the Apo-J mRNA expression levels increase from 3h group to 3d group,the top level was of 3d group. Then the levels decreased from 3d group to 7d group.(2) Comparisons between groups in the non-surgical sides:①between intracerebral hemorrhage 3d group,control group and sham-operated group,there were no differences.②Between the intracerebral hemorrhage groups,there were no differences(p>0.05).(3)Comparisons between the operated side and the non-surgical side in every model:there were no differences between the right side and the left side in control groups(p>0.05);there were differences between the operated side and the non-surgical side in sham-operated groups(p<0.05), the Apo-J mRNA level was higher in the operated side;there were significant differences between the operated side and the non-surgical side in intracerebral hemorrhage groups(p<0.001), the Apo-J mRNA level was also higher in the operated side.2. BWC levels (1) Comparison between groups in the operated sides:①between intracerebral hemorrhage 3d group,control group and sham-operated group,there was no difference between control group and sham-operated group(p>0.05). BWC level was higher in intracerebral hemorrhage 3d group than in control group and sham-operated group(p<0.001).②When comparing between intracerebral hemorrhage groups, to evaluate BWC levels by the means, he levels of the intracerebral hemorrhage groups increased from 3h to 3d,after got a 3d top,they decreased from 3d to 7d.As comparing with intracerebral hemorrhage 3d group,significant differences were got in every other intracerebral hemorrhage groups(p<0.001).(2) Comparisons between groups in the non-surgical sides:in the statistical courses of①and②,no significant overall differences were got(p>0.05).(3) Comparisons between the operated side and the non-surgical side in every model: there were no differences between the right side and the left side in control groups(p>0.05);there were differences between the operated side and the non-surgical side in sham-operated groups(p<0.05), the level was higher in the operated side;there were significant differences between the operated side and the non-surgical side in intracerebral hemorrhage groups(p<0.001 or p<0.01), the level was also higher in the operated side.3. The relation between the expression of Apo-J mRNA and BWCIn the operated sides,There was positive correlation between the Apo-J mRNA level and BWC,r=0.953(p<0.001).But in the non-surgical sides,there was negative correlation between the Apo-J mRNA level and BWC,r=-0.127(p>0.05).ConclusionsThe Apo-J mRNA expression level at the edge of the hemorrhage lesion increased. There was positive correlation between Apo-J mRNA expression and cerebral edema degree, and cerebral edema degree stands partially for brain damage.
