Objective:To investigate the expression of GDNF and GDNFmRNA in the visual central system of normal rats and monocularly deprived ambloypia rats during their development period, to offer possible evidence for probing into the mechanism of amblyopia and seeking out effective clinical methods to prevent and treat amblyopia.Methods:(1) Monocularly deprived rats model was established by suturing the eyelids of one eye of postnatal 14 days rats,and is feeded to postnatal 45 days with the control group in the same condition.The target is divided into two groups:normal group(N),monocularly deprived ambloypia opposite group(MD).(2) Use immunohistochemistry to investigate the expression of GDNF in the lateralgeniculate nucleus(LGN)and the visual cortex(V1M) of normal group(N) and monocularly deprived ambloypia opposite group(MD).(3) Use RT-PCR to investigate the expression of GDNFmRNA in the lateralgeniculate nucleus(LGN)and the visual cortex(VC) of normal group(N)and monocularly deprived ambloypia opposite group(MD).(4) microscope,photography microscope,computer image analysis,SPSS 13.0 and Independent T-test were used to get the results.Results:Result of immunohistochemistry:(1)GDNF positive cells were seen in the LGN and visual cortex of normal and monoculady deprived ambloypia rats;(2) GDNF positive cells of monocularly deprived group rats(MD) in visual cortex were significant less than that of N(p=0.013);GDNF positive cells in the LGN had no significant difference between the two groups.(P=0.822);Result of RT-PCR:(1) The expression of GDNFmRNA in the visual cortex of normal rats is higher thanthat in monocularly deprived ambloypia rats.The difference has statistical significance.Conclusion:(1)There are GDNF positive cells in the visual cortex and LGN of normal and monocularly deprived ambloypia rats.(2)The expression of GDNF positive cells in the visual cortex of monocularly deprived ambloypia rats is less that of normal rats.But there is no significant difference in the LGN.(3)The expressions of GDNF and GDNF mRNA in the visual cortex are regularity.So we presume GDNF and GDNF mRNA expression in the visual cortex is in relation to visual experience during development:abnormal experience,such as monocular deprivation leads to an downregulation of GDNF and GDNFmRNA and effect normal process of visual development.(4)The role of GDNF in visual system is possibly one of the molecular biology basis of occurance of monocularly deprived amblyopia.It perhaps as some role in the change of the visual cortex of deprived amblypia,but educe other effect in the LGN.
