Background Lung cancer is one of the most common malignant tumor in the world and is the leading cause of cancer deaths worldwide.The investigation of lung cancer indicated that more than 80%case is non-small cell lung cancer(NSCLC).Lung cancer has the poor survival rate.Only 40%-70%patient can survival 5 years even with early stage(stageâ… ,â…¡).More than 80%patient is in advanced stage or has micrometastasis when been definited.In recent times,it is important for us to use molecular method in prognosis of lung cancer and the micrometastasis.We often use fluorescence quantification PCR in our research.It is a new technology of PCR,and has combined PCR with spectral analysis,real time detection.It can improve specificity of the detection and decrease the false-positive.Objective The aim of this study was to investigate the relationship between loss of heterozygosity(LOH) of the allele genes on 3p and the diagnostic value of NSCLC.Methods A total of 32 NSCLC sample and 8 tissues of the pulmonary benign disease have analyzed by PCR.Results There were 25 of the 32 cases(78.125%) with NSCLC tumor had LOH on 3p.Among them,14 cases(43.75%) had LOH at 3p25,18 cases(56.25%) had LOH at 3p14 and 18 cases(56.25%) had LOH at 3p21.3.LOH detection in the nucleated cell in peripheral blood was 18.75%(3p25),31.25%(3p14),50%(3p21.3)and the total detection was 20 cases(65.625%).LOH detection in the nucleated cell in peripheral blood was correlation with the tumor tissue and the association detection is better than single ones.Conclusions Our analysis showed that 3p LOH occur commonly in human NSCLC.The detection of the tumor tissue and the nucleated cell in peripheral blood is beneficial to the diagnosis of NSCLC.The combined LOH data of 3 separate regions of 3p LOH is beneficial to increase the detection rate.
