Immunoglobulins(IgG),lactoferrin,Insulin-like growth factor-I,which were separated and purified from bovine colostrums,were the main bio-active components of bovine colostrum.The concentration of IL-1β,IL-1Ra,IL-4,IL-6,IL-10,IFN-γ,TNF-α,TGF-β1 in serum of rabbits,which treated with simmunoglobulins(IgG),lactoferrin,Insulin-like growth factor-I through oral administration,intraperitoneal injection,intravenous injection and the bloods of which collected at 3h after oral administration,at 2h after injection administration, respectively,was determined by a sandwich enzyme-linked immunosorbent assay(ELISA).The results clearly indicated that it was prominenced discrepancy in levels of cytokines in vivo after different administration route:The levels of proinflammatory cytokines in serum showed a marked increase after IgG oral administration and markedly decreased of anti-inflammtory cytokines,However the effect of IgG on pro-and anti-inflammtory cytokines through injection administration were adverse;The levels of anti-inflammtory cytokines in serum significantly gone up and the pro-inflammtory cytokines obviously came down after Lf through three different administration route;The levels of pro-inflammtory cytokines in serum were clearly arose after insulin-like growth factor-I oral and injection administration,and the anti-infammtory cytokines were significantly decreased.The effect of different administration on cytokines through oral and intravenous injection was less significant than the intraperitoneal injection.In this research,the rabbits' temperature was also studied as the cytokines.After IgG injection,the body temperature had great scope elevation,achieved 1.1℃,but other experimental group body temperature change within 1℃; the IgG oral administration group,the lactoferrin oral administration group,the experimental group of insulin-like growth factor-I body temperature rise within 0.25~0.5℃.According to LPS inflammation model,the thesis studied the changing regularity of pro-and anti-inflammatory cytokine in serum,and the relation of cytokines and temperature, suitable time of sampling was decided:the levels of proinflammatory cytokine declined causing the temperature to rise,along with the drop of proinflammatory cytokine levels,then the temperature gradually decreased,and the levels of anti-inflammatory cytokines rose.The pro- infammatory cytokines level in the serum rapidly increased and reached the maximum level at 1.5-2 h after injecting LPS,then gradually decreased and returned to baseline level,which the anti-inflammtory cytokines reached the maximum level at about 3 hours,The peak time of the levels of cytokines in serum through oraling LPS was one hour later than injecting LPS.Tthe bio-active components:immunoglobulin(IgG),lactoferrin,insulin-like growth factor-I which from bovine colostrums can regulate the pro-and anti-inflammtory cytokines in the serum,and then display a series of bio-function.
