Expressions Of Activin A And Its Signaling Proteins In Regenerated Liver Of Mouse Hepatectomy

Activin is a member of the transforming growth factor-beta (TGF-β) superfamily as growth and differentiation factors, not only can regulate secretion of follicle-stimulating hormone (FSH) and follistatin (FS) from anterior pituitary, but also can serve as an important regulator in the process of embryo formation, nerve differentiation, hematogenous cells proliferation, tissue repair and inflammation recovery. Activin contains three forms, activin A (βAβA), activin B (βBβB) and activin AB (βAβB). Activin A not only has the strongest effect, but also close related with liver disease. It is reported that activin can cause liver fibrosis, and the inducer of liver fibrosis, TGF-β, can also depend on the effect of endogenetic activin partially.1. Constructure of mouse hepatectomy modelExperimental animal model is useful to explore the mechanism of various diseases and drug screenings. We obtained the model of hepatectomy by excising one of three lobules mouse C57BL/6 and injected pcDNA3-ActRIP2 via vena caudalis using liposome. Whether the operation is success is depended on liver index, serum transaminase and liver tissue HE stain pathology analyse. The result shew that, the liver index were obviously higher than that of control group, p<0.05, which means liver evidently regeneration after operation; The absorben of hepatectomy group was higher than that of control based on standard curve calculater, p<0.05(ALT)and p<0.01 ( AST ) ; Pathology analyse shew that there were obviously regeneration in hepatectomy group, and the hepatoctye was bigger, the liver cell cord was derangement. From the data of above, we successfully construct liver hepatectomy regeneration model.2. RT-PCR investigate the expression of mouse liver tissue ActRⅡA,ActRⅡB mRNAThe RT-PCR result shew that ActRⅡA mRNA decreased in partial hepatectomy mouse model, p<0.01, and so that of ActRⅡB mRNA, p<0.01. The above data shew that, Activin A may take part in liver regeneration.2. The expression of Activin, FS, ActRⅡA analysed by immunohistochemistryAnti-Activin antibody immunohistochemistry stainThe anti Activin stain result shew that there were generous obviously big masculine granulaes in endochylema and nucleus in operation group, much more than control group.Anti-FS antibody immunohistochemistry stainThe anti FS stain result shew that the masculine granula is brown, and there was no obviously change between operation group and control group. Anti-ActRⅡA antibody immunohistochemistry stainThe anti ActRⅡA stain result shew that the masculine granula is brown, and was much more obviously in operation group than in control group.
