Liver Cancer is a kind of tumor that is threatening the health and lives of human kind. nowadays, the main methods of treatment are operation, radioneurosurgery and chemotherapy.Treatment and the patients after treatment results have certain limitations. RNAi can be specific to inhibition of tumor cell growth, induction of apoptosis, and other advantages, tumor gene therapy in recent years become the focus of research.This dissertation aiming at gene C-Raf conclusion the recombinant adenovirus pAd-C-Raf has been successfully constructed by the method of homogenous recombination in bacteria.Ttransfection in human hepatocarcinoma HepG-2 cell.After 48 hours,proliferation was markedly inhibited.Inhibition rate was 59.35% of cell proliferation determined by MTT assay;The growth and activity was 43.25% of HepG-2 cells explored using trypan blue staining; expression was obviously decreased determined by western blot;The inhibition rate of pAd-C-Raf recombinant adenovirus to expression of mRNA in liver cell was 42.5%.Accordingly,Recombinant adenovirus pAd-C-Raf proliferation could inhibit proliferation of human liver cancer cell and provided the basis of C-Raf gene therapy with recombinant adenovirus.
