Objective: To investigate the frequencies of the vitamin D receptor(VDR) gene Taq I ,Apa I ,Bsm I and Fok I polymorphisms in children of the Han nationality in Chengdu and reveal the relationship of VDR gene polymorphisms and growth factor, such as length,body weight,body mass index(BMI) and body surface area(BSA) of children.Methods: Five hundred and eighteen normal Han children of Chengdu were studied.Polymerase chain reaction(PCR), restriction fragment length polymorphism(RFLP) and DNA sequence analysis with restriction enzyme Taq I ,Apa I ,Bsm I and Fok I were used to analyze the VDR gene polymorphisms.Length and body weight at birth and 1-year-old were measured,and BMI and BSA were calculated.Analysis of relationship between VDR gene polymorphisms and length,body weight,BMI and BSA were made.Results: Frequencies of TT, Tt and tt genotypes were 90.7%,9.1% and 0.2%;Frequencies of T and t alleles were 95.3% and 4.7%.Frequencies of AA,Aa and aa genotypes were 10.8%,36.7% and 52.5%;Frequencies of A and a alleles were 29.2% and 70.8%.Frequencies of BB,Bb and bb genotypes were 0.4%,8.3% and91.3%;Frequencies of B and b alleles were 4.5% and 95.5%. Frequencies of FF, Ff and ff genotypes were 28.8%,48.4% and 22.8%;Frequencies ofF and f alleles were 53.0% and 47.0%. The frequencies of the four vitamin D receptor gene polymorphism sites mentioned above are similar to other nations of Mongoloid in Asia, but differ from the Caucasian.Among 199 children investigated between birth time and 1-year-old,two phenomena were found:â‘ At 1 year age,length increase of the Ff boys at Fok I polymorphism site were 1.3cm more than other types(P=0.027),but no significant differences were found in girls.â‘¡The VDR genotypic variants were not related to length,body weight,BMI and BSA at any site both at birth and 1-year-old.Conclusion: The frequencies of the vitamin D receptor gene Taq I ,Apa I ,Bsm I and Fok I polymorphisms of the Han nationality in Chengdu are similar to other nations of Mongoloid in Asia, but differ from the Caucasian.At 1 year age,average length increase of the Ff boys at Fok I polymorphism site were more higher than the FF boys.No correlation was found between the four polymorphism sites of VDR gene and length,body weight,BMI and BSA both at birth and 1-year-old.
