Objective To investigate the expression of P120 catenin in the bone marrow of a rat model for myelodysplastic syndrome and its correlation with angiogenesis of the model .Methods 34 mature Wistar-Imamichi rats, aged from 4 to 5 weeks weighing from 90 to 110g, were used in this experiment. The rats were divided into experiment group(24) and control (10). Three successive pulse doses of the chemical mutagen dimethylbenzantharcene(DMBA) were injected into the tail vein of experiment rats weekly, while the control animals received only the equivalent dose of the solvent. After establishment of the model, the expression of P120ctn in bone marrow sections was measured immunohistochemically, and the bone marrow microvessels were highlighted using anti-FVIII-related antigen staining. Both were quantified by Image-Pro Plus, and were analyzed their correlation statistically.Results The expression of P120 catenin was in stromal cells and microvessel enthelial cells of bone marrow. P120ctn localized both at cell memebrane and in cytoplasm, but not in the nucleus . P120ctn can be detected in stromal cells and microvessel endothelial cells of bone marrow. Expression of P120ctn (positive area, optical density) in control group was lower than that in the MDS group(p <0.05 or p <0.01). It showed that a significant increase in vascularity in MDS group compared with the controls (P<0.05). At the same time, the expression of P120ctn was positively related to angiogenesis in bone marrow.Conclusion The expression of P120ctn and angiogenesis were up-regulated in the model for myelodysplastic syndrome and both changes were correlated to some extent.
