This study was designed to investigate the receptor-mediated apoptosis and cell cycle specificity in cultured normal and tumour lymphocytes and the correlation between apoptosis and cell cycle.METHODS:Leukemia cell lines as of Molt-4 , Jurkat and PBL were treated by Tnf-αor Anti-fas.PBL(peripheral blood lymphocyte) cells were stimulated by phytohemagglutinin (PHA) and further incubated with Tnf-αor Anti-fas. Methods of API and Annexin V/PI are applied to illustrate the apoptosis during cell cycle progression.RESULTS:PBL cells went to apoptosis when stimulated by PHA and more under additional insults of Tnf-α, Anti-fas, while unstimulated PBL kept blunt to these insults. Molt-4 and Jurkat cells were treated by Tnf-αor Anti-fas went to apoptosis. Early G1-phase cells were determined as apoptosis by API method in PBL that stimulated by PHA. Molt-4 and Jurkat cells presented Early G1-phase pattern of cell cycle specific apoptosis under insults of Tnf-α,Anti-fas, yet.CONCLUSION:Receptor-mediated apoptosis are correlative with cell cycle and present different cell cycle specificity.
