Objective To investigate the effect of hyperthermic preconditioning on hydrogen peroxide (H2O2)-induced rat pheochromocytoma(PC12) cells cytotoxicity.Methods PC12 cells were divided into 4 groups: Control group; hyperthermic preconditioning (HPC) group; H2O2 group (cells exposed to H2O2); (HPC+ H2O2) group (cells treated with hyperthermic preconditioning before exposure to H2O2). MTT assay, LDH leakage, flow cytometry were performed to study the cytotoxicity.Results Compared with control group, MTT cell activity decreased (from 1.28±0.23 to 0.64±0.18, P<0.01), LDH leakage increased (from 82±4 to 340±20, P<0.01), and typical apoptosis was observed in the PC12 cells of H2O2 group. Compared with H2O2 group, elevation of MTT cell activity (from 0.64±0.18 to 0.89±0.10, P<0.05), reduction of LDH leakage (from 340±20 to 290±18, P<0.05) were displayed, and non-evident apoptosis was observed in (HPC+ H2O2) group.Conclusion Hyperthermic preconditioning produces protective effect against H2O2-induced PC12 cells cytotoxicity. Objective To investigate the effect of upregulation of heat shock protein 70 on PC12 cells cytotoxicity induced by hydrogen peroxide (H2O2).Methods PC12 cells were divided into 4 groups: Control group; hyperthermic preconditioning (HPC) group; H2O2 group (cells exposed to H2O2); (HPC+ H2O2) group (cells treated with hyperthermic preconditioning before exposed to H2O2). Morphology of cells was observed by taking photographs, in addition, the intracellular expression of HSP70 was detected by western blotting.Results In (HPC+ H2O2) group, the cellular morphology was markedly improved compared with H2O2 group. And the cellular HSP70 expression level in (HPC+ H2O2) group was significantly higher than that in H2O2 group. In additional experiment, the intracellular expression of HSP70 was rapidly increased at 3-6h only following HPC, and was highly maintained for at least 24-42h.Conclusion It is indicated that upregulation of heat shock protein 70 induced by HPC produces protective effect against H2O2-induced PC12 cells cytotoxicity.
