PartⅠThe cell morphology observation of mouce embryo lung fibroblast infected by Mouse Cytomegalovirus in vitroObjective To observe the cell morphology of mouce embryo lung fibroblast infected by Mouce Cytomegalovirus (MCMV) in vitro.Methods The cells infected by MCMV were observed with invert microscope and scanning electron microscope.Results With invert microscope the author could see the infected cells changed into augmentation and round .By electron microscope we could find that there were many vacuoles and lipid droplet in cell cytoplasm.And we could find the swelling mitochondrion with vacuolation and the more Golgi complex and the Endoplasmic reticulum with dilataltion. There were many MCMV particles in nucleus.Conclusion The ultrastructure of infected cells could be found to have transformed. There were many MCMV particles in nucleus after 68 hours when MCMV infected mouce embryo lung fibroblast . PartⅡThe relationship of Heat Shock Protein 70 and apoptosis induced by Mouce Cytomegalovirus in vitroObjective To study the relationship of heat shock protein 70 (HSP70) and apoptosis induced by Mouce Cytomegalovirus (MCMV) in vitro.Methods The authors detect the expression of heat shock protein 70 and MCMV-induced apoptosis of mouce embryo lung fibroblast by immunofluorescence staining and flow cytometer.Results HSP70 was strongly expressed in MCMV-infected cells after 72 hours when infected.Compared with the normal control group , the incidence of MCMV–infected apoptosis was significant.Conclusion HSP70 was induced to express in NIH3T3 cells infected by mouse cytomegalovirus(MCMV) .Meanwhile,HSP70 was expressed in cells where and when MCMV was prolifered .The apoptosis of cells could be induced by mouse cytomegalovirus . On the other hand, HSP70 could also inhibit MCMV-induce apoptosis and protect the infected cells. PartⅢThe Effect of Jinyebaidu On Heat Shock Protein 70 and apoptosis induced by Mouce Cytomegalovirus in vitroObjective To study the effect of Jinyebaidu on heat shock protein 70 (HSP70) and apoptosis induced by Mouce Cytomegalovirus (MCMV) in vitro.Methods The authors detect the expression of heat shock protein 70 and MCMV-induced apoptosis of mouce embryo lung fibroblast by immunofluorescence staining and flow cytometer.Results HSP70 was strongly expressed in MCMV-infected cells after 72 hours when infected.Compared with the normal control group , the incidence of MCMV–infected apoptosis was significant.But to jinyebaidu and gancyclovir-treated group, the incidence of apoptosis dereased.Conclusion HSP70 was induced to express in NIH3T3 cells infected by Mouse Cytomegalovirus .Meanwhile,HSP70 was expressed in cells where and when MCMV was prolifered .The apoptosis of cells could be induced by mouse cytomegalovirus . Both of jinyebaidu and gancyliovir could inhibit MCMV-induced apoptosis and resist MCMV effectively.On the other hand, HSP70 could also inhibit MCMV-induce apoptosis and protect the infected cells.
