Objective: To study the alternation of SIgA in serum, bile and intestine fluid in experimental acute intrahepatic cholestasis induced by ANIT (α-naphthyl isothiocyanate), and discuss the mechanism and the significance of the phenomena which has taken place in the intrahepatic cholestasis.Methods: 39 newborn rabbits were divided at random into four groups: 3 experimental groups (n=9) and a control group (n=12). A single dose (200mg/kg) of ANIT was administered by gavage to each experimental rabbit to induce intrahepatic cholestasis. The control group was administered with olive-oil. The specimens of serum, bile, intestine fluid and ileum tissue were collected from rabbits at 24h, 48h and 72h after intoxication respectively and detected by SIgA radioimmunologic kit. At the same time, the tissue of ileum was harvested. Spectrophotometric method was used to detect the activity of myeloperoxidase (MPO) of the ileum tissue. After the experiment, we adopted the SPSS software to analyze the significance test of the experimental results.Results:①Compared with the control group, the SIgA in serum of experimental groups were significantly increased (P<0.01) and the concentration of bile SIgA decreased dramatically (P<0.01);②There were no difference of the concentrations of SIgA in the intestine fluid between the control group and each experimental groups (P>0.05). The levels of MPO in the ileum tissues were unchanged too (P>0.05). Conclusions:①Acute intrahepatic cholestasis induced by ANIT resulted in alterations of SIgA in serum and bile, the fluctuation of the SIgA happened during the intrahepatic cholestasis is coincidence with the injury and recovery of the liver. The alternation may be used to estimate the injury of hepatobiliary system②The actual hepatobiliary system damage doesn't disturb the molecular immune of the small intestine in the short time.
