Objective: To observe stimulatory G protein expression and expression difference in Pathological scar and normal skin .To Approach stimulatory G protein expression difference and function in Pathological scar and normal skin.Methods: By operation got Pathological scar and all thickness normal skin each twenty instances from 18-38 years old scar plastic operation patients. Each Pathological scar and all thickness normal skin all from the same patient .The sample all got confirmation by HE stain. Got tissues immediately throw into nitrogen canister. Each group had 20 instances, made sign. Through RT—PCR method detected the expression of mRNA in sample. Extracted total RNA from a part of sample by TRIZOL method. Strictly acted on the operating instruction. Detected the purity of total RNA, no degradation sample ran reverse transcription, got CDNA ran PCR amplification. Electrophoresis and made use of gel image processing system analysis and took photographs. Made use of immunohistochemical method to detect the rest part of the sample. According to the operating instruction to operate. Observe distribution of expression. Distribution lied in epidermal basal cell, hypodermal fibroblast and blood capillary endothelial cell, distribution of expression in the two tissues had no difference.4,The difference of expressed in Pathological scar and normal skin epidermal basal cell had no obvious difference. The difference of expression in Pathological scar and normal skin lie in hypodermal fibroblast and blood capillary endothelial cell had obvious difference. The former is strongly positive and the latter is positive.Conclusion : 1,Pathological scar and normal skin are all express stimulatory G protein which demonstrates stimulatory G protein participates the Pathological scar and normal skin transmembrane signaling.2,Distribution lie in epidermal basal cell , hypodermal fibroblast and blood capillary endothelial cell distribution of expression in the two tissue has no difference which demonstrates stimulatory G protein participate the epidermal basal cell, fibroblast and blood capillary endothelial cell transmembrane signaling in Pathological scar and normal skin.3,The difference of expression in Pathological scar and normal skin epidermal basal cell has no obvious difference.4,The difference of expression in Pathological scar and normal skin in hypodermal fibroblast and blood capillary endothelial cell has obvious difference. Demonstrate the hypodermal fibroblast and blood capillary endothelial cell hyperplasy in Pathological scar have something to do with the high expression of stimulatory G protein.
