Objective :Three Chinese herbs lonicerae,scutellaria,forsythiae,the ingredients of Shuanghuanlian were used in anti-virus experiment in vitro ,and ribovirin to be the positive control.Exoplored those drugs effect on Madin-Darby canine kidney(MDCK) cell apoptosis induced by influenza virus A3.Methods :The Chinese herbs were separately decocted and concentrated by heating to a final concentraion of 0.50g/ml respectively . In the MDCK cell culture experiment in vitro,the cytotoxic influence of different concentrations diluted at multiple proportions was studied to determine the highest non-toxic concentration.The next diluted concentraion of the hightest non-toxic concentration was choosed to be the experiment concentration.Effects of anti-virus activity against influenza virus A3 induce MDCK cell apoptosis were estimated in terms of Cytopathic effect, Giemsa dyeing,Annexin V and PI co-dyeing flow cytometry.Result :1.The highest of nox-toxic concentration of medicaments were: lonicerae,3.90mg/ml;scutellaria,15.62mg/ml;forsythiae,15.62mg/ml.2.The morphological changes of MDCK cells virus control group were observed under the light mocroscope .It was seen the infected cells with characteristic morphological changes of apoptosis.3.The results of FCM showed that the percentage of apoptotic cells of drugs test groups (lonicerae28.13%±3.30%,scutellaria13.47%±3.04%,forsythiae 19.17%±6.55%)were obviously lower than the virus control group(P<0.001).4. The percentage of apoptotic cells of scutellaria and forsythiae test groups had no difference with drugs control groups and cells controls group(P>0.05). The lonicerae test group had difference with drugs control group and cell control group(P<0.05).5.The inhibition rate (IR)of influenza virus A3 induced MDCK cells apoptosis of scutellaria was 79.87%,IR of forsythiae was 68.67% and IR of ribavirin was 77.94%,there had no significant differences among above three groups(P>0.05).Conclusions 1. This study showed that scutellaria and forsythiae could inhibit the apoptosis of MDCK induced by influenza virus A3.2. The inhibition ratios of scutellaria,forsythiae and ribavirin had no significant differences.3.This study could not indicated Lonicerae could inhibit the apoptosis of MDCK induced by influenza virus A3 effectly.
