Objective:To explore the possibility of using intestinal submucosa acelluler matrix as cell deliverer in vaginal tissue engineering. In order to search a dependable satisfactory effect and pragmatic and natural biomaterial to provide the biomaterial supporter for vaginal tissue engineering and construct experimental model for animal research later.Method:(1)To separated the vaginal smooth muscle tissues from New Zealand female rabbits and cultured vaginal smooth muscle cells by tissue explant + collagen digestion methods in vitro.Then cultured and passaged in..Morphology of the cell monitored and its statue of growth and proliferation under the inverted microscope.Smooth muscle cells were seeded onto the matrix at density of 5.0×106/cells per cm2..The cell-matrix complex was havested at 1.2.3.And4 weeks .The growth of SMC was detemined by tissue histologicall methods .(2)To observed the surface structures of the SIS- SMC complex under the inverted microscope .The growth of SMC was determined by HE stain.( 3 ) To Detected the expression of smooth muscle a-Actin by immunohistochemical methods.Result:(1)The vaginal SMC were obtained by the methods mentioned above .The attached cells were polygon or fusiform and after confluence could be seen the marked SMC"Peak-Valley"structure in some domains..(2)The self-maded SIS was white semitransparent and ductility .No cell component could be seen in it by HE stain . (3)Specimen section werestained by HE vaginal SMCs were increasing gradually implanted form surface to internal acelluer matrix under the scanning electron microscope.(4)Specimen section were stained with the monoclonal antibody of anti-SM a-Actin by immunohistochemistry at 1,2,3and 4weeks. It was positive.Conclusion:(1)The SMC could grow.,adhere to and differentiate.on the surface of SIS and into the hollows.SIS has good biocompatibility with SMC without disturbed the cell form or inhibiting the growth of vaginal SMC,It is certificated that SIS can be used as cell deliverer in vaginal smooth muscle cells.(2)Intestinal submucosa acelluler matrix can be used as cell deliverer in vaginal tissue engineering research .It was possible to provide ideal experimental models for vaginal tissue engineering research .
