Objective: To study of the affection about pathways on the expression VEGF gene on ischemic skin flap.Methods: 30 SD rats were divided randomly into three groups: group A(pcD2 /hVEGF121, Subcutaneous n=10),group B(pcD2 /hVEGF121, artery,n=10), group C (control group,n=10),then they were received a ligation of inferior epigastric artery to induce ischemic skin flap . PcD2 /hVEGF121 were injected into the ischemic skin flap with pedicle of inferior epigastric artery through subcutaneous, artery after 7 days. RT-PCR technology and medial picture analysis system were used to detect the expression of VEGF gene.Results: compare with each group, the group A was significantly more than those of group B and group C on the expression of VEGF gene of the ischemic skin flap (p<0.01),which have significant difference. The group B was more than group C, group C was the least of all.Conclusion: The way of therapy of VEGF gene through flap subcutaneous or artery both made significant expression. The pathway through subcutaneous had higher expression of VEGF gene in ischemic skin flap than the way through artery.
