Objective: The chronic airway inflammation is many kinds of respiratory disorder,s common pathology characteristic.For example, chronic obstructive pulmonary disease (COPD),asthma,idiopathic pulmonary fibroisis(IPF) dnd so on. In recent years studies indicated, this pathology characteristic and the pulmonary alveolus macrophage and its released cytokines has the important relation.This research take COPD as an example, to observe the effect of resveratrol on cytokines released by alveolar macrophages.Chronic obstructive pulmonary disease is characterised by airflow limitation of peripheral airways that is not fully reversible and progressive and is associated with an abnormal inflammatory response of the lungs to noxious particles or gases. It causes serious morbidity and mortality and costs the global community billions of dollars per year. Alveolar macrophages are widely distributed in pulmonary alveolar . They constitute a defense line of the lung organization. Alveolar macrophages play a critical role in the pathophysiology of COPD and are a major target for future anti-inflammatory therapy. Macrophage numbers are markedly increased in the lung and alveolar space of patients with COPD.Alveolar macrophages release increased amounts of inflammatory cytokines such as interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-α), and interleukin-8 (IL-8)levels. Over-expresses inflammatory cytokines are localized to sites of alveolar destruction. The current therapy of COPD is direct to cure or cure the chronic inflammatory ,While chronic inflammation, the understanding of the basic pathogenesis of the disease is limited and there are no effective treatments. Anti- cytokines therapy has been tried with effective effect on COPD, but drugs are few. Resveratrol (3,5,4'-trihydroxystilbene) is a component of red wine extract that has anti-inflammatory and antioxidant properties and inhibits inflammatory cytokine . In this study ,we observe the effect of resveratrol on cytokines released by alveolar macrophages that were stimulated by lipopolysacharide(LPS , 10ug/ml).Methods: Alveolar macrophages were isolated from bron choalveo -larlavage fluid (BALF) from mice. The macrophages were cultured and stimulated with lipopolysaccharide (LPS , 10μg/ ml) and different concentration of resveratrol.A group was lipopolysaccharide (10mg/L). B group was LPS (10mg/L) and R (50ug/ml) . C group was LPS (10mg/L) and R(100ug/ml).D group was LPS (10mg/L) and R (150ug/ml),E group was LPS (10mg/L) and R (200ug/ml). After 4,6,12,24 hours, the IL-6,IL-8 and TNF-αwere measured by ELISA in the supernatant. Results: Result respectively as follows:(1) TNF-αlevels peaked at 6 hours, The concenttration of the TNF-αcorrespond to lower gradually in the B , C , D and E groups along with resveratrol density gradually enlargement. The TNF-αlevels in C,D and E groups were significantly lower than that of A group( P < 0. 05) . C, D, and E groups relatively did not statistical meaning. (2) IL-8 levels peaked at 8 hours, the concentration of the IL-8 correspond to lower gradually in the B , C , D and E groups along with resveratrol density gradually enlargement. The IL-8 level in E group was significantly lower than that of A group( P < 0. 05) . (3) IL-6 levels peaked at 8 hours, The concenttration of the IL-6 correspond to lower gradually in the B , C , D and E groups along with resveratrol density gradually enlargement. The IL-6 level in E group was significantly lower than that of A group( P < 0. 05) .Conclusion :Resveratrol inhibits inflammatory cytokine IL-6,IL-8 and TNF-αover-release from alveolar macrophages . In the experiment,after raising the density of the resveratrol, the level of the TNF-αhas no obvious reduce.we can think that effective dose of resveratrol be able to control this kind of cell factor. Anti-cytokines therapies are therefore needed in the future to deal with the chronic inflammation of COPD.
