Bursting activity (BA) in cerebral cortical neurons causes epileptic seizure. Seizureinduced by pentylenetetrazole (PTZ), one of the convulsantdrugs, is an excellentsystem for understanding seizure-related BA in vivo and in vitro in cultured cells.Seizure-related genes6 (SEZ-6) has been reported as a mouse brain and testis-specificgene, whose expression is increased by PTZ. The protein of SEZ-6 appears to betransmembrane proteins with long extracellular domains and short C-terminalcytoplasmic domains, possessing short consensus repeats (SCR) found in thecomplement receptor C3b/C4b. In addition, there are two domains similar to theN-terminal half of the CUB domain. These protein motifs are known to be involvedin signaling and cell-cell adhesion or recognition. To study the function of SEZ-6 andits relation to seizure has great significance.In the present study, we describe the stage-specific expression of SEZ-6 mRNAin the developing cerebellum. Immunohistochemistry showed that SEZ-6 highlyexpress in cerebellum before birth, especially in embryonic day 18, after that obviousdecline trend was observed. This is similar as the former report of SEZ-6 in cerebraltissue revealed by Northern Blots.Also is similar as the former experiments byWestern boltting.To make further study on the function of SEZ-6, we employed lipid(Lipofectamine2000TM)recombinant DNA technique, introduce expressible miceSEZ-6 gene mRNA-corresponding double-stranded DNA plasmid into primarycultured mice Purkinje neurons, then knockdown this gene on mRNA level, after thatobserve biology function of cerebellum cells. RNA interference experiences impliedthat cerebellum Purkinje cell after RNA interference have obviously apoptosis.Obviously, it seems SEZ-6 plays a role in development of cerebellum neuron.Techniques of in situ hybridization were used to analyze the cytologicaldistribution of SEZ-6 mRNA in brain tissue.We discover SEZ-6 peculiar expresses incortex and hippocampus.It is not dectected positive singal in colloid cell.Obviously, it seems SEZ-6 plays a role in development of neuron of brain.
