Abstract/Summary:
Since 1990's, as bone tissue engineering developing rapidly, the development and studies of scaffold material of bone tissue engineering has more and more been deep-going. Especially combine single material into composite materials, which is one of the hot spot in bone tissue engineering recently. In this research, first, extract wool keratin(WK) from wool, and use WK and NOCC to prepare WK/NOCC composite membrane, then characterize and analyze the composite membrane.There are two parts in this research.1. Preparation of WK solutionReduction C method was used in this part. C2H6OS was used as reduction reagent. Briefly, wool was immersed in water solution containing urea, SDS, C2H6OS. The mixture was shaken at certain temperature for certain time. Then, the mixture was filtered through stainless steel mesh. The undissolved wool was dried and weighed. The filtrate was dialyzed against degassed distilled water using cellophane tubing for 3 days. The protein concentration in the dialyzate was measured by coomassie blue dye-binding method. As the heating time and temperature increased, the dissolving rate of wool also increased, but when the mixtare was shaken at 50℃for 16h, the protein concentration was the most. So, in later research, we will use this group.These results suggest that it is possible to use Reduction C method to prepare WK solution and it bases for the next part.2. Preparation and characterization of WK/NOCC composite membraneNOCC varing in weight from 20% to 100% was used to make biocomposite membranes with WK and glycerine by solvent casting firstly. Casting temperature was respectively room temperature,37℃,50℃,70℃. Casting time was more than 48h, 20h, 15h and8h. Results showed that the pliant property of WK/NOCC composite membrane was the best and the pone rate was the lest when casting temperature was 50℃and casting time was 15h, but its mechanical properties was to be verified in next part. The structure and properties of WK/NOCC membrane were investigated through SEM,FTIR and elongation test. Molecule interactions present between WK and NOCC in the composite membrane, which allows the uniform dispersion of NOCC in WK matrix.This Contributes enhanced mechanical property and bioactivity to the composite. These results suggest that WK/NOCC composite membrane prepared in this study may serve as potential candidate of scaffold for bone tissue engineering.Based on these findings,it is possible to use WK and NOCC to make composite membrane and the composite membrane may serve as potential candidate scaffold for tissue engineering.Furthermore,we can do cytocompatibility research in the future...
