IntroductionEndometrial carcinoma is a common gynecological tumor, and one of the threekinds of female malignant tumors. With a rising incidence, it is very important incancer study. The Ras/MAPK pathway and PI3K/Akt pathway are important channelsfor the transduction of membrane receptor into the cell, and they are closely correlatedto the growth and multiplication of many kinds of tumor tissues. The recent studyshowed that there was the effect intersection between them. However, the realinteraction relationship between them has not been completely revealed yet. ERK2 andPI3K are the representative kinases of the Ras/MAPK pathway and PI3K/Akt pathwayrespectively, which are closely correlated to the cell canceration. This experimentadopted the immunohistochemical method to detect their expression in the normalendometrium tissue, endometrial carcinoma tissue and precancerosis tissue, and studiedthe relationship between their respective expression level and endometrial carcinomaclinical pathological parameter. Their correlation was studied, and their effect andinterrelationship in the occurrence and development of the endometrial carcinoma wereexplored.MethodsThe objects of this experiment were the innocent and malignant paraffin samples(totally 66) in the gynecological operation of the Department of Gynecology ofShengjing Hospital of China Medical University from 2003 to 2007. There were 43cases of endometrial carcinoma, 12 cases of atypical hyperplasia, and 11 cases of normal proliferative phase of endometrium. The expression of ERK2 and PI3K proteinwas detected in 66 cases of the innocent and malignant endometrium tissue samples bythe immtmohistochemical method (S-P method). For data analysis, SPSS11.5 softwaresystem was adopted for Ridit analysis and Spearman rank correlation analysis. WhenP<0.05, it indicated that the disparation had statistical significance.Result1. The expression of PI3K protein in endometrial carcinoma tissue, normalendometrium tissue, and atypical hyperplasia endometrium tissueThe positive expression of PI3K protein was mainly located in cytoplasm. Thepositive cells could be seen in the microscope. The cytoplasm was brown or yellow,and caryon was not dyed. The expression of PI3K protein in normal endometriumcells was mainly negative. The expression was mainly weakly positive in atypicalhyperplasia endometrium cells. The expression was chiefly strong positive andmiddle positive in endometrial carcinoma cells. Compared with each other, thethree had significant deviation (P<0.05). In different pathologically hierarchicalendometrial carcinoma tissue, the expression of PI3K protein showed no significantdifference (P>0.05). In different operation pathological staging endometrialcarcinoma tissue the expression of PI3K protein showed no significant difference(P>0.05). There was significant deviation in endometrial carcinoma tissue with orwithout nodus lymphaticus metachoresis (P<0.05).2. The expression of ERK2 protein in endometrial carcinoma tissue, normalendometrium tissue, and atypical hyperplasia endometrium tissueThe positive expression of ERK2 protein was mainly located in cytoplasm, andthere was no expression in caryon or stroma. The expression of ERK2 protein innormal endometrium cells was mainly negative. The expression was mainly weaklypositive in atypical hyperplasia endometrium cells. The expression was chieflystrong positive and middle positive in endometrial carcinoma cells. Compared witheach other, the three had significant deviation (P<0.05). In different pathologically hierarchical endometrial carcinoma tissue, the expression of ERK2 protein showedno significant difference (P>0.05). In different operation pathological stagingendometrial carcinoma tissue the expression of ERK2 protein showed no significantdifference (P>0.05). There was significant deviation in endometrial carcinomatissue with or without nodus lymphaticus metachoresis (P<0.05).3. Correlation analysis of the expression of ERK2 and PI3K protein in endometrialcarcinomaIn endometrial carcinoma tissue the expression of ERK2 and PI3K protein showedpositive correlation. The calculating coefficient correlationγwas0.3651, P<0.05.ConclusionERK2 and PI3K protein were over expressed in endometrial carcinoma tissue, andthe overexpression of PI3K protein was correlated to the lymphaticus metachoresis.Their expressions were positively correlated in endometrial carcinoma tissue. Theup-regulation of the expression of ERK2 might be related to the up-regulation of theexpression of PI3K.
