Objective:To investigate the transcription and expression of E2F3 in transitional cell carcinoma of the bladder (TCCB) and discuss their significance in carcinogenesis and progression of TCCB,as well as their relations with angiogenesis and prognosis.Methods: 1. The expression of E2F3 mRNA in sample of 34 TCCB and 19 normal bladder tissues were examined by reverse transcriptase polymerase chain reaction(RT-PCR).2. The expression of E2F3 protein in sample of 50 TCCB and 14 normal bladder tissues were examined by immunohistochemical stain.Results:1. The positive rate of E2F3 mRNA in TCCB and normal bladder tissues was 47.1%(16/34) and 0.00%(0/19)respectively.And the positive rate of E2F3 mRNA in TCCB was much higher than in normal bladder tissu(eP<0.05).There was significant difference of positive rates in different clinical stage (P<0.05)and pathological grad(eP<0.05)of TCCB. The positive rate of E2F3 mRNA in recurrent samples was 80.00%(8/10) which was significant higher than 33.33%(8/24) in primary cases (p<0.05).2. The positive rate of E2F3 protein in TCCB and normal bladder tissues was 60.00%(30/50) and 14.29%(2/14)respectively. The expression level of E2F3 protein in TCCB was much higher than in normal bladder tissue(P<0.05),at the same time it was related positively with clinical stage (P<0.05)and pathological grade(P<0.05) of TCCB.Conclusion:1. The highly positive rate of E2F3mRNA and E2F3 protein in TCCB reveals that E2F3 may play an important role in the carcinogenesis and progression of TCCB.2.The high level expression of E2F3 is related with clinical stage,pathological grade and recurrence in TCCB.It shows E2F3 may possibly be an new indicatrix for diagnosis and estimation of TCCB.3. E2F3 was possibly be an new target in gene therapy of TCCB.
