Objective : To study how to optimize on colonizationa effect of bifidobacterium to the intestinal mucosa and evaluate the antagonistic effect and pathway of bifidobacterium on radiation damage.Methods : 1) C57BL mice in SPF grade were divided into four groups from Chongqing University of Medical Sciences and evaluate the colonizationa effect of TLR2mRNA by RT-PCR and HGF by immunohistochemistry .2) C57BL mice in SPF grade were divided into bifferent groups by different ways and irradiation doses.The test of white blood cell count is finished by the different number bifidobacterium suspension coloclysis and general irradiation with 5.0 Gyγray to establish acute radiation damage pattern .The 30 survival rate is tested and reported after the bifidobacterium suspension coloclysis of 1×109cfu/ml and 8.0 gray ray to establish acute radiation damage pattern. 3) C57BL mice in SPF grade were divided into three groups. Mice in control group(n=10) were abnoml. Mice in radition damage group (n=10) and in bifidobacterium group (n=10)were received radition damage . The later were received bifidobacteria 1×109cfu/day by enteroclysm for 28 days. 24 hours latter,mice were sacrificed and collected specimen. Superoxide dismutase, catalase ,malondialdehyde in liver,TLR2mRNA in spleen ,bcl-2,NF-кB,Cytochrome C in kidney,and spine neurocyte microstruction were studied or measured.Results :1) The levels of TLR2mRNA and HGF significantly increased in enteroclysm compared to intragastric administration. 2)The number of white blood cell count significantly increased in bifidobacterium group compared to radiation damage group. The 30 survival rate increased 43% in bifidobacterium group compared to radiation damage group. 3) The levels of SOD and CAT significantly increased and MDA decreased in bifidobacterium group compared to radition damage group.Electronic microgragh of spine neurocyte and neurogliocyte showed that cytoplasm,cytoblast and plastiosome were significantly engorged ,and neurogliocyte apoptosised in earlier period. TLR2mRNA increased in C group compared to B group by RT-PCR. Cytochrome C in kidney increased in C group compared to B group, but bcl-2,NF-Кb decreased.Conclusions : Enteroclysm is better compared to intragastric administration. Bifidobacteria can degrade radition damage by scavenging free radical ,resisting membrane lipid oxidation, elevating immunity and Inducering apoptosis.So bifidobacteria possess the ability of antagonistic effect on radiation damage.
