Objective Construct and identify recombinant adenovirus mediated human tumor related gene yrdC,observe effects recombinant adenovirus on gastric cancer BGC-823 cell line. Methods The target DNA sequence of yrdC was obtained from human spleen tissue by using RT-PCR,construct high titer recombinant adenovirus Ad-yrdC by using AdEasy adenovirus carrier system,detect yrdC protein expression by using Western blot and immunohistochemical method,set up control group and transfected Ad-Null group,and study the effects Ad-yrdC on BGC-823 cell line by drawing cell growth curve and MTT chromatometry. Results Constructed high titer recombinant adenovirus mediating yrdC gene and expressed yrdC protein after transfecting BGC-823 cell line successfully,the growth velocity of transfecting Ad-yrdC group's BGC-823 cell line accelerated obviously,comparing non-transfecting group and tranfecting Ad-Null group,they had significant difference( P< 0.05);Comparing non-transfecting group,MTT value of transfecting Ad-Null group have not significant difference( P>0.05),MTT value of transfecting Ad-yrdC group is greater than transfecting Ad-Null group and non-transfecting group(P<0.05).Conclusion yrdC gene have effect that improve the growth of gastric cancer cell line obviously,the gene will become a target that gene therapy of gastric cancer.
