Penetrating keratoplasty (PKP) plays an important strategic role in ocular trauma remedy in wartime. This requests our corneal preservation capacity and mobile transportation capacity in wartime should reach to a new height. It should be settled by the construction of field eye bank. In this study, with DX mid-term medium as the preservation and transportation medium, the effect of distance transportation on viability of Corneal endothelial cells (CEC) was observed to aim to establish a firm foundation to the construction of field eye bank. In experimental study, laboratory shaker was adopted to simulate transportation condition and vibration with different transportation time and different transportation intensity was administrated to preserved corneas. And then through trypan blue combined with alizarin red staining technique, enzymohistochemical staining technique, combined with transmission electron microscopy observation, viability of CEC was detected after simulated transportation. Viability of CEC after PKP operated on rabbits was also examined. In clinical research, a retrospective study on medical records of PKP was carried out to approach if distance transportation has some influence on the effectiveness of PKP and viability of CEC. Experimental results indicated that vibration has no significant effect on viability of CEC in a limited range (effective temperature remaining in 2-6℃, sustained acceleration at 1.7m/s2 in 5 hours). Achievement ratio of PKP also has not been affected significantly. Based on these results, we concluded that DX mid-term medium can be a convenient, economic, and effective preservation and transportation medium and consistent with the essentials of field eye bank. We could apply it as groundwork to construct field eye bank which could meet the demands of peacetime and wartime.
